Heightened apoptotic priming of vascular cells across tissues and life span predisposes them to cancer therapy-induced toxicities

Johan K. E. Spetz et al.

Summary: This study reveals that vascular endothelial cells (ECs) are highly sensitive to apoptosis-inducing stress and may represent a weakest link vulnerability in the development of toxicities in multiple tissues.

Exploiting endogenous and therapy-induced apoptotic vulnerabilities in immunoglobulin light chain amyloidosis with BH3 mimetics

Cameron S. Fraser et al.

Summary: Immunoglobulin light chain (AL) amyloidosis is an incurable hematologic disorder characterized by misfolded and aggregated light chains, leading to multi-organ dysfunction. The clonal plasma cells in AL amyloidosis rely on pro-survival proteins MCL-1 and BCL-2 for their survival. The use of BH3 mimetics, such as BCL-2 inhibitors, can sensitize these cells to frontline therapies and improve treatment outcomes.

Integrated analysis of multimodal single-cell data

Yuhan Hao et al.

Summary: The study introduces a weighted-nearest neighbor analysis framework to learn the relative utility of each data type in each cell, enabling integrative analysis of multiple modalities. Applied to a CITE-seq dataset, the method constructs a multimodal reference atlas of the circulating immune system and successfully identifies and validates previously unreported lymphoid subpopulations.

Children's Oncology Group Phase III Trial of Reduced-Dose and Reduced-Volume Radiotherapy With Chemotherapy for Newly Diagnosed Average-Risk Medulloblastoma

Jeff M. Michalski et al.

Summary: In children with average-risk medulloblastoma, reducing the radiation boost volume is safe and does not compromise survival. Decreasing CSI dose in young children leads to inferior outcomes, possibly in a subgroup-dependent manner, but is associated with better neurocognitive outcome. Molecularly informed patient selection may be further explored for late-effect sparing approaches in children with MB.

Age-dependent regulation of SARS-CoV-2 cell entry genes and cell death programs correlates with COVID-19 severity

Zintis Inde et al.

Summary: The severity of novel coronavirus disease 2019 (COVID-19) varies greatly, with pediatric patients typically experiencing less severe infection than adults and the elderly. This difference may be attributed to an increase in ACE2 expression in lung epithelial cells with advancing age and the more active apoptotic defense system in younger individuals.
