Structure-guided design enables development of a hyperpolarized molecular probe for the detection of aminopeptidase N activity in vivo

Yutaro Saito et al.

Summary: Dynamic nuclear polarization (DNP) is an advanced technique that enhances the detection sensitivity of molecules in nuclear magnetic resonance (NMR)/magnetic resonance imaging (MRI). This study successfully developed a DNP-NMR molecular probe for in vivo detection of aminopeptidase N activity through precise structural investigation, calculation, biochemical assessment, and advanced molecular design.

Hyperpolarized (1-13C)Alaninamide Is a Multifunctional In Vivo Sensor of Aminopeptidase N Activity, pH, and CO2

Alice Radaelli et al.

Summary: This study demonstrates the feasibility of using spin hyperpolarization to perform real-time metabolic imaging of carbon-13-labeled substrates. It shows that hyperpolarized L-(1-13C)alaninamide can serve as a probe to detect the activity of the cell-surface tumor marker aminopeptidase-N. Furthermore, it reveals that the chemical shift of alaninamide is pH-sensitive and can be used to measure the pH levels in different compartments.

Tumor Microenvironment Biosensors for Hyperpolarized Carbon-13 Magnetic Resonance Spectroscopy

Changhua Mu et al.

Summary: Hyperpolarization of a carbon-13 molecule via dynamic nuclear polarization (DNP) using a dissolution method significantly enhances the signal-to-noise ratio of nuclear magnetic resonance (NMR) and enables the imaging of the extracellular tumor microenvironment. By studying lactate export, reduced interstitial pH, imbalanced redox equilibrium, and altered metal homeostasis in the TME through MRS, DNP-MRS shows promising outcomes for investigating features of the TME.

Evaluation of enzymatic and magnetic properties of γ-glutamyl-[1-13C]glycine and its deuteration toward longer retention of the hyperpolarized state

Yohei Kondo et al.

Summary: Dynamic nuclear polarization (DNP) is an emerging method for acquiring metabolic and physiological information in vivo. The development of a deuterated probe, gamma-Glu-[1-C-13]Gly-d(2), has been shown to extend the lifetime of hyperpolarized signals. By evaluating enzymatic conversion and magnetic properties, this study confirms the effectiveness of gamma-Glu-[1-C-13]Gly as a DNP NMR molecular probe for detecting GGT activity in serum.
