RNA N6-methyladenosine reader IGF2BP2 promotes lymphatic metastasis and epithelial-mesenchymal transition of head and neck squamous carcinoma cells via stabilizing slug mRNA in an m6A-dependent manner

Dan Yu et al.

Summary: This study reveals the oncogenic role and potential mechanism of IGF2BP2 in HNSCC, demonstrating that IGF2BP2 affects lymphatic metastasis and EMT process through the regulation of m6A modification. This suggests that IGF2BP2 may serve as a therapeutic target and prognostic biomarker for HNSCC patients with metastasis.

METTL3-mediated m6A modification of TIMP2 mRNA promotes podocyte injury in diabetic nephropathy

Ling Jiang et al.

Summary: Epigenetic changes, specifically the N-6-methyladenosine (m6A) modification, play a crucial role in diabetic nephropathy (DN). This study demonstrates that METTL3-mediated m6A modification is an important mechanism in podocyte injury in DN. Elevated METTL3 levels lead to increased inflammation and apoptosis, while METTL3 knockout alleviates these responses. Silencing METTL3 shows therapeutic potential in mitigating kidney injury and albuminuria in diabetic mice. METTL3 modulates Notch signaling through m6A modification of TIMP2, exerting pro-inflammatory and pro-apoptotic effects.

Dual role of pseudogene TMEM198B in promoting lipid metabolism and immune escape of glioma cells

Ying Zhan et al.

Summary: This study found that TMEM198B promotes the malignant progression of glioma by regulating lipid metabolism reprogramming and remodeling immune microenvironment. TMEM198B increases PLAGL2 expression, which activates ACLY and ELOVL6, influencing the behavior of glioma cells. Additionally, TMEM198B promotes M2 polarization of macrophages via glioma-derived exosomes, further facilitating immune escape of glioma cells.

Stabilization of UCA1 by N6-methyladenosine RNA methylation modification promotes colorectal cancer progression

Rong-Zhang He et al.

Summary: The study revealed that IGF2BP2 serves as a reader for m6A modified UCA1, and that METTL3 and WTAP positively regulate UCA1 expression. Mechanistically, IGF2BP2 increases the stability of m6A-modified UCA1. Additionally, IGF2BP2 is upregulated in CRC tissues compared with normal tissues, suggesting that m6A modification contributes to the upregulation of UCA1 in CRC tissues.

Silencing circPVT1 enhances radiosensitivity in non-small cell lung cancer by sponging microRNA-1208

Meifang Huang et al.

Summary: This study found that silencing circPVT1 can enhance the radiosensitivity of NSCLC cells by sponging miR-1208, inhibiting cell growth and invasion, and inducing apoptosis. It is suggested that this effect may be achieved through the activation of the PI3K/AKT/mTOR pathway.

Novel insights into the m6A-RNA methyltransferase METTL3 in cancer

Yiqing Cai et al.

Summary: N6-methyladenosine (m(6)A) is a prevalent internal RNA modification in higher eukaryotic cells, and RNA methyltransferase-like 3 (METTL3) plays a key role in this modification process. METTL3 has been shown to have pivotal roles in various human cancers, with its expression levels often associated with tumor progression and prognosis. The potential of METTL3 inhibitors in anti-tumor therapy is also discussed.
