Fifty years with aspirin and platelets

Carlo Patrono

Summary: This passage describes the significant impact of Sir John Vane's publication on the pharmacological effects of aspirin and similar drugs, as well as the subsequent discovery of the platelet arachidonic acid metabolism pathway. It also emphasizes the importance of low-dose aspirin as an antiplatelet drug.

Cyclooxygenase Inhibitors and Cancer: The Missing Pieces

Carlo Patrono

Summary: Low-dose aspirin and other cyclooxygenase inhibitors have been associated with a reduced risk of colorectal cancer and possibly other digestive tract cancers. The sequential involvement of thromboxane-dependent platelet activation and cyclooxygenase-2-driven inflammatory response in the early stages of colorectal carcinogenesis may explain these clinical benefits. This mini-review aims to analyze the evidence for a chemopreventive effect of aspirin and other cyclooxygenase inhibitors and discuss the missing pieces of this mechanistic and clinical puzzle.

Urinary Thromboxane B2 and Lethal Prostate Cancer in African American Men

Maeve Kiely et al.

Summary: In this study, we found that high levels of TXB2 were associated with prostate cancer and poor outcomes in African American men, indicating a potential role of upregulated TXA2 synthesis in promoting disease progression and metastasis. Our findings suggest a potential benefit of aspirin in preventing lethal prostate cancer through inhibition of TXA2 synthesis, especially in high-risk populations like African American men.

Inflammation and Cancer: From the Development of Personalized Indicators to Novel Therapeutic Strategies

Patrizia Ballerini et al.

Summary: Colorectal and hepatocellular carcinoma are associated with chronic inflammation, and there is a need for sensitive and specific noninvasive biomarkers for monitoring. Platelets play a role in intestinal and liver inflammation, and antiplatelet agents like Aspirin can reduce the incidence and mortality of these cancers. Platelet-mediated drug delivery systems could improve the efficacy and reduce the toxicity of anti-inflammatory agents.

The Role and Regulation of Thromboxane A2 Signaling in Cancer-Trojan Horses and Misdirection

Anthony W. Ashton et al.

Summary: The role of TXA2 signaling in cancer development and progression has been increasingly recognized, as it regulates processes such as cell growth, migration, and angiogenesis. Higher levels of TXA2 are associated with poor prognosis and metastatic disease. TXA2 signaling also modulates the tumor microenvironment and contributes to tumor formation through highly mutagenic and oncogenic by-products. Thus, targeting TXA2 signaling may be a viable approach in cancer therapy.

Summary: This study reveals the important role of platelet COX-1 in intestinal tumorigenesis in mice, by regulating cell proliferation and prostaglandins biosynthesis. Additionally, platelets contribute to the induction of COX-2 in fibroblasts, promoting the development of intestinal tumors.

Association of Thromboxane Generation With Survival in Aspirin Users and Nonusers

Summary: This study found that systemic thromboxane generation is significantly associated with all-cause and cardiovascular mortality regardless of aspirin use. In participants without cardiovascular disease, TXB2-M is associated with cardiovascular mortality, while aspirin use is associated with all-cause mortality in those with low TXB2-M levels.
