Targeting oncogenic microRNAs from the miR-371∼373 and miR-302/267 clusters in malignant germ cell tumours causes growth inhibition through cell cycle disruption

Testicular germ cell tumour cells release microRNA-containing extracellular vesicles that induce phenotypic and genotypic changes in cells of the tumour microenvironment

Summary: Malignant germ-cell-tumours (GCTs) are characterized by dysregulation of microRNA (miRNA/miR-), specifically overexpression of miR-371, miR-373, and miR-302/367 clusters. These miRNAs are released in extracellular vesicles (EVs) and may serve as potential biomarkers. This study comprehensively investigated the role of EVs and their miRNA cargo in the tumor microenvironment (TME) of testicular GCTs (TGCTs). The results showed that TGCT cells secreted EVs containing miR-371, miR-373, and miR-302/367 miRNAs. Co-culture of TGCT cells with TME cells resulted in increased levels of these miRNAs in TME cells. Fluorescent labeling demonstrated the internalization of TGCT-derived EVs by TME cells. Treatment of TME cells with EVs from different TGCT subtypes led to increased miRNA levels. Overexpression of miR-371a in TME cells resulted in functional changes, including collagen contraction and angiogenesis. These findings suggest that TGCT cells communicate with nontumor stromal TME cells through EVs enriched with oncogenic miRNAs, potentially contributing to tumor progression.
