Summary: Traumatic damage to the spinal cord often leads to permanent tissue defects. We have discovered that injecting imidazole-poly(organophosphazene) hydrogel (I-5) can bridge cystic cavities and promote the formation of newly assembled extracellular matrix (ECM). However, the ECM created by the hydrogel contains components that can hinder axonal growth. To address this issue, we have developed a dual-functional system by equipping the hydrogel with an additional enzyme, ARSB, which can degrade the hindering components. This complex has shown promising results in promoting axonal growth and improving locomotor recovery in spinal cord regeneration.

The up-regulation of the RhoA/ROCK pathway contributes to neuronal apoptosis, neuroinflammation, blood-brain barrier dysfunction, astrogliosis, and axon growth inhibition in IS and SCI. Inhibiting the Rho/ROCK pathway with miRNAs alleviates apoptosis, neuroinflammation, oxidative stress, and axon growth inhibition in IS and SCI. Further studies are required to explore the significance of ncRNAs in IS and SCI and to establish new strategies for preventing and treating these devastating diseases.
