Prefrontal microglia deficiency during adolescence disrupts adult cognitive functions and synaptic structures: A follow-up study in female mice

Prenatal opioid exposure inhibits microglial sculpting of the dopamine system selectively in adolescent male offspring

Summary: The current opioid epidemic has led to an increase in the number of children exposed to opioids during prenatal development, and this exposure has been linked to social and cognitive abnormalities in young adulthood. However, the mechanisms behind these effects are not well understood. This study demonstrates that microglia, the immune cells of the brain, play a crucial role in translating prenatal opioid exposure into changes in neural systems and behavior. Maternal oxycodone self-administration during pregnancy was found to reduce microglial phagocytosis of dopamine D1 receptors in the brain, leading to higher receptor density and behavioral deficits in adult male offspring.

Prenatal environmental stressors impair postnatal microglia function and adult behavior in males

Summary: Gestational exposure to environmental toxins and socioeconomic stressors is linked to neurodevelopmental disorders in males. We conducted a study on mice and found that only male offspring displayed lasting behavioral abnormalities and changes in brain networks related to social interactions. Prenatal stressors were found to affect the function of microglial cells in the anterior cingulate cortex during early postnatal development in males. Inhibiting microglial phagocytosis in the same brain region of wild-type mice during the critical period mimicked the impacts of prenatal stressors on male-specific behavior, suggesting that environmental stress alters neural circuit formation in males by impairing microglial function during development.
