Echocardiographic evaluation of regurgitant fraction in dogs with subclinical myxomatous mitral valve disease: Method comparison, effects of pimobendan, and reproducibility

Summary: This study evaluated the effectiveness of Simpson's method and Motion-mode in estimating regurgitant fraction (RF) in dogs with subclinical myxomatous mitral valve disease (MMVD), as well as the effects of pimobendan and the reproducibility of the measurement methods. The results showed that both Simpson's method and Motion-mode had a curvilinear relationship with left atrium-to-aortic root ratio and varied considerably across different stages of the disease. Pimobendan significantly reduced RF, and RF SMOD_TSV may be a more reliable technique for quantifying RF.

Vericiguat preserved cardiac function and mitochondrial quality in a rat model of mitral regurgitation

Summary: The study aimed to investigate the effects of the sGC stimulator Vericiguat on cardiac function, blood pressure, cardiac mitochondrial quality, and fibrosis in rat models of chronic mitral regurgitation. The results showed that Vericiguat preserved cardiac function and structure and significantly reduced systolic blood pressure. It also improved mitochondrial quality by reducing ROS production, depolarization, and swelling, and decreased fibrosis area.

Pharmacokinetics of pimobendan following oral administration to New Zealand White rabbits (Oryctolagus cuniculus)

Summary: This study aimed to investigate the pharmacokinetics and potential adverse effects of pimobendan after oral administration in New Zealand White rabbits. The results showed that the half-life of pimobendan in rabbits was 3.54 hours, and its plasma concentration could be detected for up to 24 hours. The active metabolite, ODMP, could be detected in rabbits for 24 to 36 hours.

Preliminary Bioequivalence of an Oral Pimobendan Solution Formulation with Reference Solution Formulation in Beagle Dogs

Summary: This study compared the pharmacokinetic characteristics, bioequivalence, and cardiovascular effects of a custom-made oral pimobendan solution (PS) formulation with a reference solution (RS) formulation in conscious, healthy dogs. The results showed that both formulations had comparable plasma concentrations of pimobendan and its active metabolite, and exhibited bioequivalence. In addition, both formulations significantly increased cardiac contraction with no difference in contractility between them. The PS formulation can be used as an alternative to the RS formulation for the management of congestive heart disease.
