IL-33 priming amplifies ATP-mediated mast cell cytokine production

Summary: Inflammatory responses are necessary for blocking pathogen infection but can also lead to hypersensitivity and chronic inflammation. This study found that IL-33 enhances the ATP-induced cytokine production in mast cells, potentially through the P2X7 receptor and calcineurin/NFAT pathway. Furthermore, IL-33 + ATP co-stimulation enhances the recruitment of eosinophils and macrophages.

They are related to other leukocyte subsets but form a separate branch in the hematopoietic system. Mast cells share some functions with the nervous system and express unique neuronal markers. Genes hyper-expressed in mast cells, but largely inactive in other myelocytes, include those related to traffic/lysosomes, the dopamine system, Ca2+-related entities, adhesion molecules, and transcription factors.

NINJ1 protein plays an essential role in inducing PMR, with Ninj1-deficient macrophages showing impaired release of intracellular proteins and distinctive ballooned morphology. This study overturns the long-held belief that cell death-related PMR is a passive event, highlighting the active role of NINJ1 in mediating PMR.
