Combining genetic engineering and bioprocess concepts for improved phenylpropanoid production

Discovery of Novel Tyrosine Ammonia Lyases for the Enzymatic Synthesis of p-Coumaric Acid

Summary: p-Coumaric acid (p-CA) is a key precursor for flavonoid biosynthesis, and tyrosine ammonia lyases (TALs) catalyze the synthesis of p-CA from l-tyrosine. In this study, 875 putative TALs and 46 putative phenylalanine/tyrosine ammonia lyases (PTALs) were analyzed, and 5 TALs and 3 PTALs were characterized and found to exhibit the proposed enzymatic activity. The TAL from Chryseobacterium luteum sp. nov (TAL(clu)) showed the highest affinity and conversion efficiency towards l-tyrosine. Optimization of reaction conditions resulted in p-CA yields of 2.03 g/L after 8 hours by TAL(clu) and 2.35 g/L after 24 hours by TAL from Rivularia sp. PCC 7116 (TAL(rpc)). These TALs are potential candidates for the production of flavonoid precursor p-CA using whole-cell systems.

Simple and efficient enzymatic procedure for p-coumaric acid synthesis: Complete bioconversion and biocatalyst recycling under alkaline condition

Summary: p-Coumaric acid, a natural active substance with antioxidant, anticancer, and other properties, can be biologically produced via one-step deamination of L-tyrosine by tyrosine ammonia-lyase (TAL). In this study, a new highly active TAL (Fc-TAL) from Flavobacterium columnare was identified and immobilized on silica nanospheres for enhanced stability and recyclability. The immobilized TAL showed higher stability and retained over 50% activity even after 10 consecutive catalytic cycles. Furthermore, the immobilized enzyme achieved a high conversion rate and yield in the biosynthesis of p-coumaric acid under alkaline condition.

The number of catalytic cycles in an enzyme's lifetime and why it matters to metabolic engineering

Summary: Metabolic engineering utilizes enzymes to construct biosystems for specific tasks, with the working life and failure modes of enzymes being important indicators of engineering performance. By adopting catalytic cycles until replacement (CCR) as a metric, the functional lifespan of enzymes in vivo can be calculated, with enzymes in different organisms showing varying CCR values. Enzymes susceptible to damage by specific substrates, products, or mechanisms tend to have shorter lifespans, suggesting that enzyme engineering to increase CCR and reduce replacement costs could be beneficial.
