Protein adsorption and separation with monomodal and multimodal anion exchange chromatography resins. Part II. Mechanisms of protein aggregation on the chromatographic surface

Joey A. Roberts et al.

Summary: The study found that protein oligomers are formed directly on the surface of multimodal resins and are eluted as discrete species with increasing salt concentration. The formation of aggregates is influenced by factors such as incubation time, pH, and salt concentration, leading to the formation of irreversibly bound species. Higher pH and lower salt concentration in load buffers, as well as higher temperatures, promote aggregation, while minimal aggregation occurs for Nuvia HP-Q.

Evaluation of lentiviral vector stability and development of ion exchange purification processes

Ronit Ghosh et al.

Summary: In this study, a high yield, HCP clearance anion exchange capture process for lentiviral vector (LVV) purification was developed through parallel batch stability and chromatographic screens in combination with linear and step gradient experiments. By screening and evaluating resin performance under stable conditions, optimal operating steps for LVV bioprocessing were established to achieve high yield and protein clearance.

Advances in Lentivirus Purification

Ana Sofia Moreira et al.

Summary: Lentiviral vectors (LVs) are increasingly used in gene and cell therapies due to their ability to integrate the genome in both dividing and nondividing cells. The production and purification processes have evolved significantly, but the demand for higher purity LVs is driving the development of new materials and strategies. Large-scale LV manufacturing still relies on adherent cell culture, with the industry moving towards suspension cultures, and purification strategies mainly involve batch chromatography and membrane technology. New solutions are being developed to enhance production schemes and expand clinical applications.
