Structural characterization and hepatoprotective activity of exopolysaccharide from Bacillus velezensis SN-1

Summary: This study isolated a fraction of released exopolysaccharide (rEPS) from Bacillus velezensis SN-1 and characterized it. The results showed that rEPS-2 has hepatoprotective effects and can enhance antioxidant capacity by regulating the expression of antioxidant genes and activating antioxidant enzymes. Furthermore, rEPS-2 can influence and modulate the gut microbiota of mice with liver injury caused by carbon tetrachloride (CCl4).

Rational engineering of cofactor specificity of glutamate dehydrogenase for poly-?-glutamic acid synthesis in Bacillus licheniformis

Summary: This study presents a novel approach for improving gamma-PGA production by switching the cofactor specificity of glutamate dehydrogenase. The researchers used structural modeling and molecular docking to predict the binding modes of NADH and NADPH, and obtained site-specific mutants to alter cofactor specificity. The mutant RocGD276E showed the highest increase in gamma-PGA yield and decreased the yields of main by-products. Enzymatic properties confirmed that this mutant had higher affinity for NADH, shifting the coenzyme preference from NADPH to NADH while maintaining catalytic efficiency comparable with the wild-type.
