MALDI Imaging Assisted Discovery of a Di-O-glycosyltransferase from Platycodon grandiflorum Root

Weiwei Tang et al.

Summary: A matrix-assisted laser desorption/ionization mass spectrometry imaging (MALDI MSI) assisted genome mining strategy was applied to discover a glycosyltransferase (GT) named PgGT1 from the root of Platycodon grandiflorum. PgGT1 was found to catalyze the synthesis of platycoside E (PE) by sequentially attaching two beta-1,6-linked glucosyl residues to the glucosyl residue at the C3 position of platycodin D (PD). The study also revealed the crucial roles of residues S273, E274, and H350 in stabilizing the sugar donor and positioning the glucose for optimal glycosylation. These findings provide insights into the biosynthetic pathway of PE and have potential implications in industrial biotransformation.

Monitoring Direct Thrombin Inhibitors With Calibrated Diluted Thrombin Time vs Activated Partial Thromboplastin Time in Pediatric Patients

Rida A. Hasan et al.

Summary: The plasma diluted thrombin time (dTT) showed a more stable dose-response relationship and stronger correlation with DTI dose compared to activated partial thromboplastin time (aPTT) in pediatric patients receiving argatroban and bivalirudin. The variation in dose-response curves was higher for argatroban compared to bivalirudin.

Inhibitory activities of flavonoids from Scutellaria baicalensis Georgi on amyloid aggregation related to type 2 diabetes and the possible structural requirements for polyphenol in inhibiting the nucleation phase of hIAPP aggregation

Ying Wang et al.

Summary: The active compounds in Scutellaria baicalensis can effectively inhibit hIAPP aggregation, with baicalein and baicalin, which have ortho-hydroxyl groups on the Aring, being more effective. Flavonoids with ortho-hydroxyl groups on the B-ring also exhibit high anti-nucleation activity. These results provide a valuable structural template for the development of polyphenol-based inhibitors of amyloid peptides.

Aptamer-functionalized metal-organic frameworks (MOFs) for biosensing

Mengzhen Lv et al.

Summary: MOFs, as a class of crystalline porous materials, have been widely applied in various fields due to their nanoscale framework structure, adjustable pore size, large specific surface area, and good chemical stability. With the functionalization of aptamers, MOFs-based biosensors have shown great potential in medical diagnostics, food safety inspection, and environmental surveillance. Researchers are also exploring stimuli-responsive MOFs and the integration of MOFs on microfluidic devices for sensing applications.

A review of in vitro studies of the anti-platelet potential of citrus fruit flavonoids

Beata Olas

Summary: Despite being rich sources of flavonoids that have the potential to modulate blood platelet activity, citrus fruits have mainly been studied in vitro, with relatively weak evidence regarding their antiplatelet activity and poorly understood mechanisms. Additionally, the concentrations of these flavonoids studied in vitro do not always correspond with their physiological concentrations in vivo, and citrus fruit flavonoids are characterized by low bioavailability. Further detailed studies on the antiplatelet potential of citrus flavonoids, especially in in vivo models, are needed.
