Multifunctional rotational active valve for flow control in paper-based microfluidic devices

Gohar Hussain et al.

Summary: In this paper, a multifunctional, simple, robust and cost-effective rotational active valve (RAV) for paper-based microfluidics devices is presented. The RAV achieves variable flow velocity and has been demonstrated in various applications such as sequential delivery, multi-reagent valve regulation, and fluid distribution. A multistep biochemical assay for the detection of total protein, uric acid, and albumin protein in human serum has also been successfully conducted using the RAV. The authors believe that the RAV has the potential to be commercialized as a cost-effective and reliable solution for complex biochemical assays in paper-based devices, enabling point-of-care diagnostic platforms.

Multiplexed Ultrasensitive Sample-to-Answer RT-LAMP Chip for the Identification of SARS-CoV-2 and Influenza Viruses

Minsun Song et al.

Summary: Prompt on-site diagnosis of SARS-CoV-2 with other respiratory infections will have minimized the global impact of the COVID-19 pandemic through rapid, effective management. However, no such multiplex point-of-care (POC) chip has satisfied a suitable sensitivity of gold-standard nucleic acid amplification tests (NAATs). It presents a rapid multiplexed ultrasensitive sample-to-answer loop-mediated isothermal amplification (MUSAL) chip operated by simple LED-driven photothermal amplification to detect six targets from single-swab sampling.

Droplet digital recombinase polymerase amplification for multiplexed detection of human coronavirus

Ji Wook Choi et al.

Summary: Since the outbreak of COVID-19, detection technologies in molecular diagnosis applications, particularly droplet digital PCR (ddPCR), have gained significant attention. The combination of isothermal amplification strategies with ddPCR has made it a popular method for on-site testing. However, non-specific amplification still poses a challenge. In this study, a multiplexed droplet digital recombinase polymerase amplification (MddRPA) assay was developed, offering rapid detection, precise quantification, and multiplexing capability for viral infections.

Microfluidics Technology in SARS-CoV-2 Diagnosis and Beyond: A Systematic Review

Mohd. Raeed Jamiruddin et al.

Summary: This review explores the application of microfluidic technologies in COVID-19 pandemic, particularly their combination with different detection techniques for rapid and sensitive detection of virus antibodies, antigens, and nucleic acids. The study found that polymers like PDMS and PMMA are commonly used materials for microfluidic devices, with centrifugal force being the most widely used fluid manipulation technique. Different types of detection techniques were employed depending on the target, such as spectrometer-based detection for antibody detection and electrochemical-based detection for antigen detection. Sensitivity and specificity of the devices were found to vary with the combination of detection-based technique and fluid manipulation technique. Most microfluidic devices are simple and can be easily deployed in various environments, playing a significant role in improving infection detection rate and healthcare services.
