The application of antidepressant drugs in cancer treatment

Yunxi Zheng et al.

Summary: Antidepressants are psychotropic drugs used to treat mental illness. They have been found to have anticarcinogenic effects, affecting various signaling pathways and the microenvironment. Mechanisms include apoptosis, antiproliferative effects, oxidative stress, DNA damage, immune response and inflammation, and inhibition of multidrug resistance. This review summarizes the impacts and mechanisms of 10 antidepressants in preventing cancer and discusses their potential drawbacks. Further research is needed for a full understanding and to explore new avenues for cancer prevention and treatment.

Occludin facilitates tumour angiogenesis in bladder cancer by regulating IL8/STAT3 through STAT4

Fan Yang et al.

Summary: According to recent studies, the tight junction protein occludin (OCLN) is highly expressed in bladder cancer tissues and is associated with a poor prognosis. Downregulation of OCLN inhibits tumor angiogenesis, while overexpression of OCLN has the opposite effect. Mechanistically, OCLN increases levels of interleukin-8 (IL8) and phosphorylated Signal Transducer and Activator of Transcription 3 (p-STAT3) to promote bladder cancer angiogenesis. OCLN regulates IL8 transcriptional activity through the transcription factor STAT4.

DNA damage and NF-κB inactivation implicate glycyrrhizic acid-induced G1 phase arrest in hepatocellular carcinoma cells

Wei-Shu Wang et al.

Summary: Glycyrrhizic acid plays an important role in anti-HCC therapy by inducing DNA damage, apoptosis, and expression of cell cycle regulatory proteins to inhibit the growth of HCC cells. Moreover, it can induce G(1) phase arrest in HCC cells and suppress relevant signaling pathways.

Activation of the 5-HT7 receptor and MMP-9 signaling module in the hippocampal CA1 region is necessary for the development of depressive-like behavior

Monika Bijata et al.

Summary: This study investigates the impact of 5-HT7R/MMP-9 signaling and dendritic spine plasticity on major depressive disorder. The results show that activation of 5-HT7R and chronic stress both negatively affect dendritic spine elongation and density in the hippocampus, which are associated with depressive-like behavior.

7-Epitaxol induces apoptosis in cisplatin-resistant head and neck squamous cell carcinoma via suppression of AKT and MAPK signalling

Hui-Ju Yang et al.

Summary: This study investigated the anticancer efficacy of a major paclitaxel metabolite called 7-Epitaxol in cisplatin-resistant HNSCC and found that it exhibits cytotoxic effects by inducing cell cycle arrest and apoptotic pathways. The study also revealed that 7-Epitaxol can suppress AKT and MAPK signaling pathways to induce cancer cell apoptosis.

NF-KB inhibitors gifted by nature: The anticancer promise of polyphenol compounds

Chengcheng Guan et al.

Summary: Polyphenol compounds, as natural antioxidants, have anti-inflammatory and antioxidant properties, and are believed to have various medicinal benefits for improving human health. Their anticancer effect is mainly attributed to their strong antioxidant and immunomodulatory effects. By targeting NF-KB, polyphenol compounds can exert antioxidant and immunomodulatory effects, hindering the occurrence and development of tumors. They have advantages over conventional anticancer therapies as they have minimal side effects and can attenuate the toxic effects of other treatments, showing great potential in cancer prevention and treatment.

Pre-clinical investigation of STAT3 pathway in bladder cancer: Paving the way for clinical translation

Sepideh Mirzaei et al.

Summary: Effective cancer therapy requires the identification of signaling networks, with STAT3 playing a significant role in promoting proliferation, invasion, chemoresistance, and metastasis in bladder cancer cells. Inhibiting STAT3 may enhance chemotherapy efficacy by overcoming chemoresistance in BC. Multiple molecular pathways and non-coding RNAs act as upstream regulators of STAT3 in BC cells.

Suppression of EGFR/PKC-δ/NF-κB Signaling Associated With Imipramine-Inhibited Progression of Non-Small Cell Lung Cancer

Po-Fu Yueh et al.

Summary: The study found that imipramine effectively induces apoptosis of NSCLC cells by suppressing the EGFR/PKC-d/NF-κB pathway, leading to increased DNA damage and decreased invasion and migration potential. In both in vitro and in vivo models, imipramine demonstrated tumor growth inhibition similar to the standard therapy erlotinib (EGFR inhibitor). Non-obvious body weight loss and liver pathology changes were observed in mice treated with imipramine.

In vitro antimetastatic activity of Momordica balsamina crude acetone extract in HT-29 human colon cancer cells

Karabo Serala et al.

Summary: The study demonstrated that the crude acetone extract of Momordica balsamina (MBE) inhibited the metastatic ability of HT-29 colorectal cancer (CRC) cells in vitro. MBE consists of a mixture of compounds with cytotoxic effects against HT-29 cells and can suppress reactive oxygen species formation, cell invasion, migration, and adhesion. The antimetastatic effects of MBE were associated with the regulation of various proteins related to cancer metastasis.
