Establishment of an experimental model of canine malignant mesothelioma organoid culture using a three-dimensional culture method

Yomogi Sato et al.

Summary: Due to limited studies on canine malignant mesothelioma (cMM), the pathogenesis of the disease and effective treatment options are not well understood. This study successfully generated cMM organoids for the first time using pleural effusion samples. RNA sequencing analysis revealed upregulation of cell adhesion molecule pathways and increased expression of E-cadherin in cMM organoids compared to 2D cultured cells. These cMM organoids could serve as a valuable experimental tool for studying MM therapy in dogs and humans.

Organoid Models for Infectious Disease

Sarah E. Blutt et al.

Summary: Infectious diseases have significant impacts on individuals and society. The use of pluripotent and tissue stem cell-derived organoids as models for studying human infections allows researchers to recapitulate disease characteristics and gain new insights into host-microbe interactions. Increasing the complexity of organoid cultures can further improve their application as infection models.

Distribution of histopathologic types of primary pulmonary neoplasia in dogs and outcome of affected dogs: 340 cases (2010-2019)

Jourdan B. McPhetridge et al.

Summary: This study provides updated information on the distribution of histopathologic types of primary pulmonary neoplasia in dogs and evaluates the effect of postoperative adjuvant chemotherapy in dogs with pulmonary carcinoma. Pulmonary carcinoma was the most frequently encountered tumor type, and there were significant differences in median survival time between dogs with pulmonary carcinomas, dogs with histiocytic sarcomas, and dogs with neuroendocrine tumors. However, there were no significant differences in median survival time between dogs with pulmonary carcinomas that did and did not receive adjuvant chemotherapy.

Lapatinib as first-line treatment for muscle-invasive urothelial carcinoma in dogs

Shingo Maeda et al.

Summary: This study describes the therapeutic efficacy of lapatinib as a first-line treatment for muscle-invasive urothelial carcinoma. The results show that dogs treated with lapatinib/piroxicam had a greater reduction in the size of the primary tumor and improved survival compared to those treated with piroxicam alone. Exploratory analyses also showed that HER2 overexpression was associated with response and survival in dogs treated with lapatinib.

The Obscure Potential of AHNAK2

Mohamed Zardab et al.

Summary: AHNAK2 is a newly discovered protein with a strong association with oncogenesis in various epithelial cancers. It has a tripartite structure and is believed to be involved in the formation of large multi-protein complexes. High expression of AHNAK2 is found in several cancers, including clear cell renal carcinoma and pancreatic ductal adenocarcinoma, and is associated with poor prognosis. Limited research has been conducted on the function and relation of AHNAK2 with cancer, but early studies suggest its potential as a future biomarker and therapeutic target.

AHNAK2 is a biomarker and a potential therapeutic target of adenocarcinomas

Meng Xu et al.

Summary: This study investigates biomarkers and therapeutic targets for cervical adenocarcinoma. The researchers find that AHNAK2 is highly expressed in cervical adenocarcinoma and other adenocarcinoma tissues, but not in normal gland tissues. In vitro and in vivo experiments show that AHNAK2 knockdown significantly inhibits cell proliferation, migration, tumor progression, and metastasis of adenocarcinomas.

Summary: This study aims to develop a new research model for pet cancers by generating a 2.5D organoid culture model directly from cancer-diseased dogs and cats using special media. The direct 2.5D organoids showed improved attachment, growth, marker expression, and proliferation speed compared to those cultured in normal media. Additionally, the direct 2.5D organoids exhibited concentration-dependent responses to anti-cancer drugs, with different sensitivity profiles among the strains.

Summary: The global cancer burden in 2020 saw an estimated 19.3 million new cancer cases and almost 10.0 million cancer deaths. Female breast cancer surpassed lung cancer as the most commonly diagnosed cancer, while lung cancer remained the leading cause of cancer death. These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.
