iNOS contributes to heart failure with preserved ejection fraction through mitochondrial dysfunction and Akt S-nitrosylation

Yongzheng Guo et al.

Summary: This study investigated the role of inducible nitric oxide synthase (iNOS) and its mechanisms in a HFpEF mouse model induced by high-fat diet and Nx-nitro-L-arginine methyl ester. Both pharmacological inhibition and iNOS deficiency improved mitochondrial dysfunction, oxidative stress, and Akt S-nitrosylation, leading to ameliorated HFpEF phenotype in mice. These findings suggest that iNOS inhibition may be a potential therapeutic strategy for HFpEF.

Identification of a novel native peptide derived from 60S ribosomal protein L23a that translationally regulates p53 to reduce myocardial ischemia-reperfusion

Anwen Yin et al.

Summary: The novel native peptide PDRL23A has been identified as a potential treatment for myocardial ischemia-reperfusion (I/R) injury, as it can effectively alleviate hypoxia-induced cardiomyocyte injury and improve mitochondrial function and redox homeostasis.

SIRT3-AMPK signaling pathway as a protective target in endothelial dysfunction of early sepsis

Huilin Yu et al.

Summary: This study found that the SIRT3-AMPK signaling pathway plays an important role in inhibiting vascular inflammation and endothelial dysfunction during early sepsis. Activation of AMPK can reverse the reduction of eNOS expression, prevent endothelial dysfunction, and improve survival. Inhibition of AMPK has opposite effects.

Cyanocobalamin prevents cardiomyopathy in type 1 diabetes by modulating oxidative stress and DNMT-SOCS1/3-IGF-1 signaling

Masao Kakoki et al.

Summary: Kakoki et al. found that cyanocobalamin (B12) can prevent cardiomyopathy in type 1 diabetic mice by reducing ROS levels and restoring IGF-1 levels through DNMT-SOCS1/3 signaling. B12 shows promising cardioprotective effects and may serve as an option to prevent diabetic cardiomyopathy.

Tubeimoside I Ameliorates Myocardial Ischemia-Reperfusion Injury through SIRT3-Dependent Regulation of Oxidative Stress and Apoptosis

Dingyi Lv et al.

Summary: The study demonstrates that TBM can protect against MIRI through regulation of oxidative stress and apoptosis, and this effect is dependent on SIRT3. TBM may be a potential drug candidate for MIRI treatment.

Mitochondrial ROS promote mitochondrial dysfunction and inflammation in ischemic acute kidney injury by disrupting TFAM-mediated mtDNA maintenance

Meng Zhao et al.

Summary: Reducing mtROS levels attenuated renal dysfunction, mitochondrial damage, and inflammation in IRI-AKI mice. The loss of TFAM can impact mtDNA maintenance, mitochondrial energy metabolism, and cytokine release. Insights into the role of mtROS and TFAM in IRI-AKI provide potential therapeutic targets for renal repair.
