STIM1 is a core trigger of airway smooth muscle remodeling and hyperresponsiveness in asthma

Martin T. Johnson et al.

Summary: This study reveals the up-regulation of STIM1 in the airway smooth muscle of asthmatic mice, which is involved in the metabolic and transcriptional reprogramming process of airway remodeling and the mechanism of airway hyperresponsiveness. STIM1 regulates the activation and secretion of airway smooth muscle cells, increases mitochondrial mass, oxidative phosphorylation, and glycolytic flux, and drives the development of asthma. It is proposed as a potential target for asthma therapy.

IL-37 protects against airway remodeling by reversing bronchial epithelial-mesenchymal transition via IL-24 signaling pathway in chronic asthma

Kang-ni Feng et al.

Summary: This study investigated the effect of IL-24 on EMT and the potential of IL-37 to mitigate IL-24-induced EMT in chronic asthma. Results showed that IL-24 promoted cell migration and EMT biomarker expression via the STAT3 and ERK1/2 pathways, while IL-37 partially reversed these effects by blocking the same pathways. In vivo experiments demonstrated that IL-24 silencing or IL-37 treatment could reverse EMT biomarker expression in a mouse model of chronic asthma.

Mast cell-derived serotonin enhances methacholine-induced airway hyperresponsiveness in house dust mite-induced experimental asthma

Erika Mendez-Enriquez et al.

Summary: This study found that mast cells release serotonin, which enhances bronchoconstriction and AHR. Mast cells play a key role in the pathogenesis of asthma, and M3-directed treatments may also act by targeting mast cells.

Airway smooth muscle pathophysiology in asthma

Summary: Airway smooth muscle (ASM) cells play a central role in the pathogenesis of asthma, controlling muscle tone and participating in airway hyperresponsiveness and inflammation. Therapeutic approaches for asthma require a comprehensive understanding of ASM structure and function.
