Epigenetic reprogramming in cancer: From diagnosis to treatment

Pedro Mikael da Silva Costa et al.

Summary: Disruption of the epigenetic program of gene expression is a hallmark of cancer, which is related to tumor heterogeneity, self-renewal, and differentiation. The reversible nature of epigenetic modifications provides a promising approach for cancer treatment by restoring the cancer epigenome through the inhibition of epigenetic modifiers. These alterations can also serve as biomarkers for early cancer diagnosis, and there are already approved epigenetic therapies for cancer treatment.

Transcriptome analysis of newly established carboplatin-resistant ovarian cancer cell model reveals genes shared by drug resistance and drug-induced EMT

Juran Kralj et al.

Summary: A novel OC cell model was established, and two transcriptome analysis approaches identified TMEM200A and PRKAR1B as important genes in carboplatin resistance and metastasis, with high predictive and prognostic values for OC patients. Compared to parental cells, carboplatin-resistant cells showed increased sensitivity to PI3K/Akt/mTOR pathway inhibitors and enhanced sensitivity to carboplatin when treated with a transcription inhibitor.

Mutational drivers of cancer cell migration and invasion

Nikita M. Novikov et al.

Summary: Genomic instability and mutations play a crucial role in cancer development by affecting cell proliferation, apoptosis, and immune response; mutations are also associated with cancer metastasis, a significant event in cancer progression and a life-threatening issue for patients; despite skepticism, increasing evidence suggests that mutations may trigger and accelerate cancer cell migration and invasion.

EMT-associated microRNAs and their roles in cancer stemness and drug resistance

Guangtao Pan et al.

Summary: EMT is involved in various malignant behaviors of cancers such as proliferation, invasion, and metastasis; miRNAs play a crucial role in regulating EMT phenotype and influencing cancer stemness and drug resistance; understanding the relationship between EMT-associated miRNAs and cancer stemness/drug resistance is beneficial for both basic research and clinical treatment.

Could MicroRNAs Be Useful Tools to Improve the Diagnosis and Treatment of Rare Gynecological Cancers? A Brief Overview

Riccardo Di Fiore et al.

Summary: Gynecological cancers are a significant public health issue with high incidence among females. While rare gynecological cancers often have poor prognosis and risk delayed diagnosis, there is growing interest in using miRNAs to improve diagnosis, therapy and prognosis, although research on rare gynecological cancers is limited.

Ovarian cancer: epigenetics, drug resistance, and progression

Weiwei Xie et al.

Summary: Ovarian cancer is associated with activation of oncogenes and inactivation of tumor suppressor genes, with epigenetic processes playing a key role in tumorigenesis. MicroRNA and long non-coding RNA are promising molecular markers for precision medicine in OC. Clinical application of epigenetic approaches is currently limited by hurdles such as heterogeneity of OC and insufficient sample sizes, but holds promise for future clinical investigation into diagnosis, treatment, and prognosis.

Epigenetic Alterations and Mechanisms That Drive Resistance to Targeted Cancer Therapies

Narendra Wajapeyee et al.

Summary: Cancer cells harbor multiple genetic and epigenetic alterations, but targeted therapy can still effectively treat various types of cancer. Resistance often emerges, posing a major clinical hurdle. Mechanisms for resistance to targeted therapy have been identified through alterations to the transcriptome and chromatin regulatory proteins.

The dynamic broad epigenetic (H3K4me3, H3K27ac) domain as a mark of essential genes

Tasnim H. Beacon et al.

Summary: The broad epigenetic domain, marked by H3K4me3, is associated with cell identity, essential cell functions, and the metastatic potential of cancer cells. It interacts with various histone modifications and forms a hub of interactions involving transcription factors, enhancers, and super-enhancers. This dynamic chromatin structure supports frequent transcription bursts and may act as a transcription factory.

The Roles of the Let-7 Family of MicroRNAs in the Regulation of Cancer Stemness

Yuxi Ma et al.

Summary: Cancer stem-like cells play a critical role in tumorigenesis, cancer progression, treatment resistance, and metastatic relapse. The let-7 family of microRNAs is important in regulating cancer stemness through various RNA-binding proteins, long non-coding RNAs, and circular RNAs. Targeting let-7 for cancer therapeutics has the potential to improve treatment efficacy by specifically targeting CSCs and modulating metabolic pathways.
