Reverse Transcriptase Inhibition Disrupts Repeat Element Life Cycle in Colorectal Cancer

Summary: Aberrant RNA expression of repetitive sequences and retrotransposition are common in colorectal cancer and can be targeted by the nucleoside reverse transcriptase inhibitor 3TC. The preferential effect of 3TC on p53-mutant cell lines is attributed to direct binding of p53 to repeat elements. Clinical trials have shown that single-agent 3TC treatment benefits a subset of metastatic colorectal cancer patients. 3TC induces the accumulation of immunogenic RNA:DNA hybrids, interferon response, and DNA damage response in colorectal cancer tumorspheres. Epigenetic and DNA-damaging agents enhance the cytotoxicity of 3TC. These findings highlight the therapeutic potential of targeting the viral mimicry of repeat elements in colorectal cancer.
