A Transformable Amphiphilic and Block Polymer-Dendron Conjugate for Enhanced Tumor Penetration and Retention with Cellular Homeostasis Perturbation via Membrane Flow

Lei Gu et al.

Summary: Efficient penetration and retention of therapeutic agents in tumor tissues can be achieved through rational design of drug delivery systems. In this study, a polymer-dendron conjugate is proposed, which undergoes a structural transformation triggered by a specific enzyme, leading to the release of therapeutic agents and interaction with tumor cell membranes for deep penetration and prolonged retention in the tumor tissues. Additionally, this conjugate disrupts cell homeostasis, making tumor cells more susceptible to photodynamic therapy.

A Self-Degradable Conjugated Polymer for Photodynamic Therapy with Reliable Postoperative Safety

Hongye Huang et al.

Summary: In this study, a self-degradable type-I photosensitizer based on conjugated polymers was developed, which can effectively generate superoxide radicals under light irradiation and further degrade into nontoxic micro-molecules. This strategy helps to address the phototoxicity issues of residual photosensitizers and accelerate the metabolism of conjugated polymers, enhancing postoperative safety.

Photosensitizing drug reactions

Simone Montgomery et al.

Summary: Photosensitizing drug reactions are skin eruptions that occur in patients using photosensitizing medications after exposure to ultraviolet radiation. These reactions can be classified into phototoxic and photoallergic reactions, with difficulties in distinguishing between the two, but management is similar and prevention is key.

Emerging Strategies in Enhancing Singlet Oxygen Generation of Nano-Photosensitizers Toward Advanced Phototherapy

Mohammad Tavakkoli Yaraki et al.

Summary: This paper reviews the recent development of nano-photosensitizers and nanoplasmonic strategies in photodynamic therapy. Firstly, the limitations of classical photosensitizers in PDT are discussed, followed by an introduction to three types of new generation nano-photosensitizers and their synthesis methods. The strategies of using metal nanoparticles to enhance singlet oxygen generation efficiency of nano-photosensitizers are also discussed. Lastly, the future prospects of these nanoengineering strategies are highlighted.

Metabolizable Photosensitizer with Aggregation-Induced Emission for Photodynamic Therapy

Wenbo Wu et al.

Summary: A highly efficient metabolizable type-II photosensitizer with aggregation-induced emission characteristics was developed in this study, demonstrating good anti-tumor PDT efficiency in mouse models. Importantly, the photosensitizer can be gradually oxidized by endogenous hypochlorite after PDT and subsequently excreted through urine within 24 hours of administration.
