Multi-scale Modelling of Shear Stress on the Syncytiotrophoblast: Could Maternal Blood Flow Impact Placental Function Across Gestation?

Tet Chuan Lee et al.

Summary: The placenta plays a critical role in fetal development and is influenced by shear stress exerted by maternal blood flow. This study found that the syncytiotrophoblast expresses mechanosensitive proteins and motor proteins associated with cilia, with higher expression levels in early pregnancy compared to term. Computational models of blood flow at different scales showed that shear stress on the syncytiotrophoblast is higher in early pregnancy. These findings suggest that blood flow and shear stress have important effects on placental development and function from early in pregnancy.

How trophoblasts fuse: an in-depth look into placental syncytiotrophoblast formation

Stephen J. Renaud et al.

Summary: In humans, cell fusion is restricted to only a few cell types under normal conditions. In the placenta, cell fusion is a critical process for generating syncytiotrophoblast and plays a crucial role in ensuring pregnancy health.

Endothelial shear stress signal transduction and atherogenesis: From mechanisms to therapeutics

Lei He et al.

Summary: Atherosclerotic vascular disease and its complications are leading causes of mortality globally. Studies have shown that endothelial cells are influenced by low or disturbed shear stress at curvatures and bifurcations in the arterial tree, playing a role in the formation and development of atherosclerotic plaques. Understanding mechanosensitive signaling cascades may offer new translational strategies for therapeutic intervention of atherosclerotic vascular disease.

Notch signaling and fluid shear stress in regulating osteogenic differentiation

Yuwen Zhao et al.

Summary: Osteoporosis is a common bone and metabolic disease characterized by bone density loss and microstructural degeneration. This study investigates the regulatory role of Notch signaling during osteogenic differentiation of human bone marrow-derived mesenchymal stem cells (hMSCs) exposed to physiologically relevant fluid shear stress using a locked nucleic acid (LNA)/DNA nanobiosensor. The results provide evidence that shear stress regulates osteogenic differentiation through Notch signaling.

The exploration of pluripotency space: Charting cell state transitions in peri-implantation development

Martin F. Pera et al.

Summary: Recent studies of pluripotent stem cells have identified an intermediate stage called the formative state between naive pluripotency and primed pluripotency, which has important implications for deriving human embryo models.

Molecular characteristics of established trophoblast-derived cell lines

Jana Pastuschek et al.

Summary: This study aims to evaluate molecular markers and features matching different trophoblast subpopulations in trophoblastic cell lines to provide guidance on their suitability and relevance for specific research questions. The results suggest that characteristics and markers of choriocarcinoma-derived cell lines are more similar to in vivo trophoblast than immortalized cell lines, making them potentially more suitable models for research on trophoblast biology and pathology.
