Molecular Approaches to Protein Dimerization: Opportunities for Supramolecular Chemistry

Dung Thanh Dang

Summary: Protein dimerization plays a crucial role in many biological processes, and understanding its regulation is important for biomedical applications. The limitations of engineered dimerization provide opportunities for molecular chemistry to induce protein dimerization. This review discusses molecular control over protein dimerization and activation, highlighting both established and novel approaches.

Dimeric translationally controlled tumor protein-binding peptide 2 attenuates imiquimod-induced psoriatic inflammation through induction of regulatory T cells

Hyunsoo Cho et al.

Summary: This study found that dTCTP dysregulation in psoriasis leads to immune imbalance through the suppression of regulatory T cells (Treg). The use of dTCTP inhibitor dTBP2 can maintain immune homeostasis and alleviate inflammatory skin diseases.

PEGylation improves the therapeutic potential of dimerized translationally controlled tumor protein blocking peptide in ovalbumin-induced mouse model of airway inflammation

Hyeran Seo et al.

Summary: In this study, the researchers chemically conjugated dTBP2 with 10 kDa polyethylene glycol (PEG) to improve its therapeutic potential. The modified PEG-dTBP2 showed higher inhibitory action against airway inflammation in mice compared to the unmodified dTBP2. The results suggest that PEG-dTBP2 could be considered as a potential candidate drug for regulating allergic inflammation.

Role of regulatory T cells in psoriasis pathogenesis and treatment

L. Nussbaum et al.

Summary: Psoriasis is a chronic inflammatory disease with a strong genetic component that can be triggered by environmental factors. Tregs play a fundamental role in immune homeostasis and contribute to the prevention of autoimmune disease by suppressing immune responses, but their functional regulation remains unclear in the context of psoriasis.

Blockade of translationally controlled tumor protein attenuated the aggressiveness of fibroblast-like synoviocytes and ameliorated collagen-induced arthritis

Mingyo Kim et al.

Summary: The study demonstrates the pivotal role of HRF/TCTP in the pathogenesis of rheumatoid arthritis (RA), with HRF/TCTP significantly associated with inflammatory responses and joint destruction. Inhibiting HRF/TCTP with dimerized HRF/TCTP-binding peptide-2 (dTBP2) shows promising therapeutic effects in RA patients and collagen-induced arthritis (CIA) mice, highlighting HRF/TCTP as a potential novel biomarker and therapeutic target for the diagnosis and treatment of RA.

Allergic Inflammation Caused by Dimerized Translationally Controlled Tumor Protein is Attenuated by Cardamonin

Haejun Pyun et al.

Summary: The study demonstrated that cardamonin can exert anti-allergic and anti-inflammatory effects by interacting with dTCTP, reducing secretion of IL-8, inhibiting cell migration and IgE production, as well as reducing bronchial remodeling. Cardamonin also showed potential in therapy of allergic diseases by inhibiting NF-kappa B activity.
