Calcipotriol abrogates cancer-associated fibroblast-derived IL-8-mediated oxaliplatin resistance in gastric cancer cells via blocking PI3K/Akt signaling

Zhen-Xiong Zhao et al.

Summary: This study confirms the specific localization of VDR in gastric cancer tissues and demonstrates that activation of VDR can suppress CAF-induced chemotherapy resistance in gastric cancer. The study also reveals that CAF-derived IL-8 induces oxaliplatin resistance through activating the PI3K/AKT pathway, and treatment with vitamin D can attenuate the tumor-supportive effect of IL-8 on gastric cancer cells.

Curcumin inhibits the cancer-associated fibroblast-derived chemoresistance of gastric cancer through the suppression of the JAK/STAT3 signaling pathway

In-Hye Ham et al.

Summary: This study aimed to investigate the role of the JAK/STAT3 signaling pathway in the chemoresistance of gastric cancer induced by cancer-associated fibroblasts (CAF). The results showed that CAFs increased the resistance of gastric cancer cells to chemotherapy, and this was associated with the activation of the JAK/STAT3 pathway. However, the natural product curcumin was found to inhibit the activation of this pathway and sensitize the cancer cells to chemotherapy. These findings suggest that targeting the JAK/STAT3 pathway may be a potential strategy to overcome chemoresistance in gastric cancer, and curcumin could be a promising natural compound for this purpose.

TGF-β-Induced FLRT3 Attenuation Is Essential for Cancer-Associated Fibroblast-Mediated Epithelial-Mesenchymal Transition in Colorectal Cancer

Mengdi Yang et al.

Summary: Cancer-associated fibroblasts (CAF) play a role in the progression of colorectal cancer. This study shows that CAFs can downregulate FLRT3 expression through the activation of the TGF-beta/SMAD4 signaling pathway, promoting epithelial-mesenchymal transition (EMT) and the invasive behavior of colorectal cancer cells. LY2109761, a dual inhibitor of TGF-beta receptor type I and II, can inhibit metastasis and may be a potential treatment option for advanced colorectal cancer.

mRNA Subtype of Cancer-Associated Fibroblasts Significantly Affects Key Characteristics of Head and Neck Cancer Cells

Barbora Peltanova et al.

Summary: This study delves into the interaction between cancer-associated fibroblasts (CAFs) and head and neck cancer cells, illustrating their impact on the metabolic state, mechanobiology, and aggressiveness of cancer cells. The findings suggest that the growth and progression of tumors require the right collaboration between cancer cells and CAFs, with specific mRNA subtypes playing a crucial role in supporting primary or metastatic cancer cells. Furthermore, the research indicates that the progression of HNSCC may be influenced by the support of CAFs in the tumor microenvironment, and the heterogeneity of this disease may be attributed to the level of cooperation between CAFs and epithelial cancer cells.

The prognostic and therapeutic implications of distinct patterns of argininosuccinate synthase 1 (ASS1) and arginase-2 (ARG2) expression by cancer cells and tumor stroma in non-small-cell lung cancer

Alexandra Giatromanolaki et al.

Summary: ARG2 and ASS1 enzymes are extensively expressed in NSCLC stroma and cancer cells, respectively. Auxotrophic tumors have a poor prognosis, potentially by utilizing Arg, thus reducing Arg-dependent TIL anti-tumor activity. ASS1 expression in cancer cells would allow Arg fueling of (iNOS+)TILs and enhance anti-tumor immunity. However, upregulation of ARG2 in CAFs may divert Arg from TILs, allowing immune escape. Identification of these three distinct phenotypes may be useful in the individualization of Arg-targeting therapies and immunotherapy.
