Summary: The study identifies a high affinity interaction between SVEP1 and the orphan receptor PEAR1, which promotes phosphorylation of PEAR1 and activation of AKT/mTOR signaling in vascular cells and platelets. Mice lacking SVEP1 exhibit reduced platelet activation, while exogenous SVEP1 induces PEAR1-dependent activation of platelets. Targeting this receptor-ligand interaction may offer a potential therapeutic strategy for cardiovascular and thrombotic diseases.

Summary: The study reveals a novel role for SVEP1 and the integrins alpha 4 beta 1 and alpha 9 beta 1 in reducing VSMC contractility. Inhibition of these proteins significantly enhances calcium levels in VSMCs and leads to increased vessel contraction, mediated via VGCCs, PKC, and Rho A/Rho kinase dependent mechanisms.

Summary: This study investigated the role of integrin alpha 9 beta 1 in the development of atherosclerosis. The results showed that depletion of integrin alpha 9 beta 1 in VSMCs or myeloid cells did not affect the formation of atherosclerotic plaque. This suggests the presence of other potential receptors or alternative cell types expressing integrin alpha 9 beta 1 in the development of atherosclerosis.
