In vitro reconstitution of calcium-dependent recruitment of the human ESCRT machinery in lysosomal membrane repair

Summary: The endosomal sorting complex required for transport (ESCRT) machinery plays a key role in repairing damaged plasma and lysosome membranes. Calcium ion (Ca2+) signaling is shown to be involved in ESCRT recruitment to damaged sites. This study demonstrates that the calcium-binding regulatory protein ALG-2 directly binds to negatively charged membranes in a Ca2+-dependent manner, recruiting ALIX and orchestrating the assembly of downstream repair components.

Researchers developed a model to simulate the depolymerization process and found that changes in filament composition can trigger membrane deformation, leading to the release of cargo-loaded vesicles.

Design principles of the ESCRT-III Vps24-Vps2 module

Summary: Our previous studies on ESCRT-III subunits Snf7 and Vps24 revealed the association between the helix-4 region of Snf7 and the helix-1 region of Vps24. Mutations in the helix-1 region of another ESCRT-III subunit Vps2 were found to functionally replace Vps24 in Saccharomyces cerevisiae, revealing essential features of both subunits. This research proposes three minimal features necessary for ESCRT-III function - spiral formation, lateral association of spirals via heteropolymerization, and binding to the AAA + ATPase Vps4 for dynamic remodeling.
