Targeting G-quadruplexes to achieve antiviral activity

Emanuela Ruggiero et al.

Summary: With the emergence and fast mutation rates of new viruses, there is a need for new antiviral targets and compounds. Most existing antiviral drugs target only a few specific viruses by affecting viral nucleic acid processing. However, selectively targeting nucleic acids of interest is challenging. Recently, G-quadruplexes (G4s), which are alternative structures to the classic double helix, have attracted attention due to their important biological roles. Molecules that selectively target G4s have been developed and investigated as potential antiviral drugs for various human pathologies, including viral infections.

Queenie Fernandes et al.

Summary: The emergence of novel and evolving variants of SARS-CoV-2 has created the need for newer and more adaptive diagnostic methods. However, developing rapid and sensitive diagnostic technologies has become more challenging due to emerging variants and varying symptoms. Vaccines and drugs are essential for prevention and protection, and constant development is underway. This review discusses the challenges posed by novel variants and the evolution of diagnostic techniques, as well as the development, mechanisms, advantages, and drawbacks of vaccines and drugs and their immunological impact.

Anti-tumoural activity of the G-quadruplex ligand pyridostatin against BRCA1/2-deficient tumours

Florian J. Groelly et al.

Summary: The compound pyridostatin shows high specificity against BRCA1/2-deficient tumors and tumors resistant to PARP inhibitors. It disrupts replication leading to DNA double-stranded breaks and can be repaired in the absence of BRCA1/2 by non-homologous end joining. Additionally, pyridostatin triggers immune responses and can be further potentiated by combining it with paclitaxel. Overall, pyridostatin has potential for therapeutic development in cancer patients with BRCA1/2 mutations.

A phase 2a clinical trial of molnupiravir in patients with COVID-19 shows accelerated SARS-CoV-2 RNA clearance and elimination of infectious virus

William A. Fischer et al.

Summary: In a clinical trial, the safety, tolerability, and antiviral efficacy of molnupiravir were evaluated in unvaccinated individuals with confirmed SARS-CoV-2 infection. The study found that participants receiving high-dose molnupiravir had a shorter time to viral RNA clearance and a lower detection rate of infectious virus. Molnupiravir was well tolerated across all doses.

SARS-CoV-2 Mutations and Their Impact on Diagnostics, Therapeutics and Vaccines

Suresh Thakur et al.

Summary: With the global increase in COVID-19 infections, the emergence of SARS-CoV-2 variants was inevitable. Mutations, especially in the spike protein, have been found to alter the transmissibility and virulence of the virus. The WHO has classified the variants with fitness-enhancing mutations into variants of concern, interest or under monitoring. These variants pose a significant threat as they exhibit higher transmissibility and immune evasion. This article reviews the mutational landscape of SARS-CoV-2 proteins and their impact on diagnostics, therapeutics, and vaccines, as well as the effectiveness of approved vaccines, antibody therapy, and convalescent plasma against the currently prevalent variants.

RNA helicase DDX5 enables STAT1 mRNA translation and interferon signalling in hepatitis B virus replicating hepatocytes

Jiazeng Sun et al.

Summary: The study demonstrates that RNA helicase DDX5 plays a crucial role in regulating STAT1 translation by resolving a G-quadruplex structure in the 5' UTR of STAT1 mRNA, and is essential for IFN response. The absence of DDX5 correlates with reduced STAT1 expression and weakened anti-viral effect of IFN-alpha in HBV-infected cells, highlighting the importance of DDX5 in controlling the dynamic range of IFN response.

RNA G-quadruplex formed in SARS-CoV-2 used for COVID-19 treatment in animal models

Geng Qin et al.

Summary: The study reveals the presence of highly conservative and stable G-quadruplexes (G4s) in SARS-CoV-2, which play a dual role in inhibiting viral replication and transcription processes. The cationic porphyrin compound TMPyP4, targeting SARS-CoV-2 G4s, shows excellent antiviral activity and effectively suppresses SARS-CoV-2 infection in animal models. This research provides a novel therapeutic approach by targeting the secondary structures within the SARS-CoV-2 genome and opens new possibilities for the design of selective drug candidates.
