Hydropersulfides inhibit lipid peroxidation and ferroptosis by scavenging radicals

Uladzimir Barayeu et al.

Summary: Ferroptosis is a type of cell death caused by radical-driven lipid peroxidation. Our study shows that sulfane sulfur species, particularly hydropersulfides, scavenge free radicals and suppress ferroptosis. Cysteine can support ferroptosis resistance independently of the GPX4 pathway by providing sulfur for S-0 biosynthesis.

Andrographolide Inhibits Static Mechanical Pressure-Induced Intervertebral Disc Degeneration via the MAPK/Nrf2/HO-1 Pathway

Cunxin Zhang et al.

Summary: This study investigates the molecular mechanism of andrographolide (ADR) in inhibiting static mechanical pressure-induced apoptosis in nucleus pulposus cells (NPCs) and its role in inhibiting intervertebral disk degeneration (IDD). The findings suggest that ADR inhibits apoptosis and reactive oxygen species (ROS) accumulation in NPCs, improves cell viability, and suppresses IDD through the activation of the OK/Nrf2/HO-1 signaling pathway.

Multiple myeloma cells induce lipolysis in adipocytes and uptake fatty acids through fatty acid transporter proteins

Cristina Panaroni et al.

Summary: This study reveals that adipocytes in the bone marrow play a role in supporting multiple myeloma cells and that the lipolysis pathway is involved in this interaction. The uptake of free fatty acids by myeloma cells promotes their proliferation, while higher concentrations of fatty acids induce lipotoxicity. Blocking the uptake of fatty acids by myeloma cells could be a potential therapeutic strategy for multiple myeloma.

CircRNA-ST6GALNAC6 increases the sensitivity of bladder cancer cells to erastin-induced ferroptosis by regulating the HSPB1/P38 axis

Li Wang et al.

Summary: This study describes a regulatory mechanism for ferroptosis sensitivity in bladder cancer that is dependent on the expression levels of circST6GALNAC6. CircST6GALNAC6 inhibits HSPB1 and promotes cell ferroptosis by blocking the phosphorylation site of HSPB1 and activating the P38 MAPK signaling pathway. Enhancing the expression of circST6GALNAC6 or using it as a biomarker for ferroptosis sensitivity is significant for the development and application of ferroptosis intervention methods in bladder cancer.

Hydropersulfides Inhibit Lipid Peroxidation and Protect Cells from Ferroptosis

Zijun Wu et al.

Summary: Hydropersulfides (RSSH) play important roles in vivo as scavengers of oxidants and electrophiles. They are highly reactive towards phospholipid-derived peroxyl radicals and can effectively inhibit ferroptosis. The reactivity of RSSH can be improved by designing specific precursors. RSSH and their precursors have been shown to inhibit ferroptosis in cell culture.
