Engineering protein-based therapeutics through structural and chemical design

Sasha B. Ebrahimi et al.

Summary: Protein-based therapeutics have revolutionized disease treatment, with proteins projected to be among the top selling drugs in the near future. This review discusses the design strategies that have transformed protein-based structures into effective therapeutics and highlights emerging protein engineering strategies. Such strategies aim to enhance the stability, targetability, cell permeability, and reduce immunogenicity of proteins, paving the way for improved therapeutic properties.

Oxidation degree of a cell membrane model and its response to structural changes, a coarse-grained molecular dynamics approach

Hugo Aceves-Luna et al.

Summary: This study used molecular dynamics simulation to observe the effect of different percentages of oxidized lipids on the structure of Dioleoylphosphatidylcholine membranes. The results showed that higher concentrations of oxidized lipids led to more damage to the membrane, as evidenced by increased lipid area, decreased thickness and membrane packing, and decreased hydrophobicity within the membrane.

Role and Posttranslational Regulation of Cx46 Hemichannels and Gap Junction Channels in the Eye Lens

Mauricio A. Retamal et al.

Summary: Connexins are proteins that can form hemichannels and gap junction channels. In the lens, connexins play important roles in metabolic cooperation between cells and their modulation is involved in cataract formation. This review focuses on the functions of Cx46 hemichannels and gap junction channels and their posttranslational modifications.

Molecular dynamics simulations of the effects of lipid oxidation on the permeability of cell membranes

Daniel Wiczew et al.

Summary: The sustained permeability of cell membranes in electroporation phenomenon may be caused by membrane lesions that underwent secondary oxidation. Molecular dynamics simulations and free energy calculations demonstrate that these lesions provide a better model to explain the post-pulse permeability and conductivity of electropermeabilized cells. These findings are further discussed in relation to sonoporation and ferroptosis, suggesting substantial lipid oxidation might be triggered in similar processes.

Connexin hemichannels regulate redox potential via metabolite exchange and protect lens against cellular oxidative damage

Yumeng Quan et al.

Summary: Our study reveals that Cx43 hemichannels (HCs) in lens epithelial cells play a crucial role in protecting the lens against oxidative stress by facilitating the exchange of oxidants and antioxidants, thereby maintaining redox homeostasis. Activation of Cx43 HCs by oxidative stress results in a reduction of intracellular reactive oxygen species (ROS) accumulation and regulation of anti-oxidative gene expression, contributing to preventing cataract formation during aging.

Connexin Hemichannel Activation by S-Nitrosoglutathione Synergizes Strongly with Photodynamic Therapy Potentiating Anti-Tumor Bystander Killing

Chiara Nardin et al.

Summary: The study investigated bystander effects induced by photodynamic therapy in murine melanoma models, revealing that photoactivation triggers cell death pathways in both irradiated and bystander tumor cells. Hemichannel activity and ATP release play crucial roles in inducing bystander cell death, and combination treatment with S-nitrosoglutathione enhances this effect significantly.
