Regioselective syntheses of 2-oxopyridine carbonitrile derivatives and evaluation for antihyperglycemic and antioxidant potential

Summary: A library of 2-oxopyridine carbonitriles 1-34 was synthesized by regioselective nucleophilic substitution reactions. The synthetic compounds exhibited potential antidiabetic and antioxidant activities. Compound 1 showed the highest inhibitory activity against α-glucosidase, while compound 6 showed the highest inhibitory activity against α-amylase. The compounds followed a competitive mode of inhibition, and molecular docking studies revealed their interaction profiles with the target enzymes.

Dicyanoanilines as potential and dual inhibitors of α-amylase and α-glucosidase enzymes: Synthesis, characterization, in vitro, in silico, and kinetics studies

Summary: The study focuses on the synthesis of dicyanoaniline derivatives and their inhibition activities against alpha-amylase and alpha-glucosidase enzymes. Among the synthesized compounds, five of them showed excellent inhibition against the enzymes. Enzyme kinetic studies revealed different types of inhibition mechanisms, and computer simulations demonstrated the involvement of these molecules in binding interactions at the enzyme's active site.
