Phoma spp. an untapped treasure of cytotoxic compounds: current status and perspectives

Exploration of Potent Cytotoxic Molecules from Fungi in Recent Past to Discover Plausible Anticancer Scaffolds

Summary: Fungi can produce various scaffolds with unique biological activities, but so far, no molecule derived from fungi has become an anti-cancer drug on the market. Every year, numerous cytotoxic molecules of fungal origin are published, making it necessary to critically analyze these compounds to identify the potent ones. A review published in 2014 mentioned the best cytotoxic fungal metabolites and their status in drug development. In this report, we included 176 cytotoxic molecules isolated from fungi after 2014 and categorized them based on their potencies. Emphasis was given to discuss the 42 molecules that showed IC50 values below 1 mu M. This review provides potent fungal scaffolds that can be prioritized as drug candidates for cancer therapeutics.

Summary: In this study, five previously undescribed oblongolides were isolated from Phoma bellidis Neerg., along with four known compounds. The structures and absolute configurations of these compounds were determined using various experimental methods. Phomaone A was identified as the first example of oblongolides with glycol directly linked by two C-C bonds. The cytotoxicity of the compounds was evaluated against human cancer cell lines, showing significant activity except for phomaone A.

Summary: Two new pyranone derivatives and a new coumarin, along with other known compounds, were isolated from the endophytic fungus Phoma sp. YN02-P-3, which was obtained from a Paulownia tree in Yunnan, China. These newly identified compounds showed moderate inhibitory activity against human tumor cells.

Undescribed diphenyl ethers betaethrins A-I from a desert plant endophytic strain of the fungus Phoma betae AB Frank (Didymellaceae)

Summary: Ten diphenyl ethers, including nine undescribed analogs, were isolated from the desert plant endophytic fungus Phoma betae. The structures of these compounds, named betaethrins A-I, were determined using NMR, HR-ESI-MS, and X-ray diffraction experiments. It was found that the B-ring had a shielding effect on H-6 in certain analogs, leading to differences in the 1H-NMR chemical shift values. These compounds exhibited strong antioxidant activities and moderate cytotoxicity against cancer cell lines. The study highlights the importance of exploring endophytic fungi from unique environments for the discovery of bioactive metabolites.
