Engineering Escherichia coli for a high yield of 1,3-propanediol near the theoretical maximum through chromosomal integration and gene deletion

Nonthaporn Wong et al.

Summary: In this study, a highly efficient 1,3-propanediol-producing E. coli strain NSK015 was developed by integrating heterologous genes into the chromosomal DNA of E. coli. The developed strain demonstrated high 1,3-propanediol production using glycerol and cassava starch as co-substrates, without the need for plasmid expression systems, antibiotics, IPTG, and rich nutrients.

D-Lactic Acid Production from Sugarcane Bagasse by Genetically Engineered Saccharomyces cerevisiae

Warasirin Sornlek et al.

Summary: In this study, the production of D-lactic acid (D-LA) was improved in Saccharomyces cerevisiae by gene editing and conventional breeding. By deleting specific genes and crossbreeding with acid-tolerant yeast strains, the production and yield of D-LA were significantly increased, while the production of glycerol and ethanol were minimized.

Mechanisms underlying lactic acid tolerance and its influence on lactic acid production in Saccharomyces cerevisiae

Arne Peetermans et al.

Summary: One of the main challenges in lactic acid production through microbial fermentation is the negative impact of lactic acid accumulation on the producing cells. This review extensively discusses how Saccharomyces cerevisiae addresses these issues and explores different strategies, both targeted and non-targeted, to improve lactic acid tolerance.

Systematic engineering of Saccharomyces cerevisiae for D-lactic acid production with near theoretical yield

Akaraphol Watcharawipas et al.

Summary: This research successfully engineered Saccharomyces cerevisiae to produce D-lactic acid from glucose at near theoretical yield, representing progress towards efficient microbial production of D-lactic acid. The strain ASc-d789M produced D-lactic acid at a titer of 17.09 g/L in shake-flasks and 40.03 g/L in fed-batch fermentation, demonstrating high productivity and potential for industrial applications.

Growth and autolysis of the kefir yeast Kluyveromyces marxianus in lactate culture

Shou-Chen Lo et al.

Summary: The study found that Kluyveromyces marxianus grown with lactate as the sole carbon source showed optimal growth and released protein under different pH conditions. Lactate induced the autolysis of K. marxianus, leading to the release of proteins and peptides. These findings are crucial for future probiotic and kefir studies involving K. marxianus.
