Machine-learning methods applied to integrated transcriptomic data from bovine blastocysts and elongating conceptuses to identify genes predictive of embryonic competence

Maria Belen Rabaglino et al.

Summary: This study developed a gene signature model to predict embryonic competence for survival by integrating transcriptomic data from blastocysts and elongating conceptuses. The potential biomarkers were validated with independent embryonic data sets using machine-learning algorithms. The results showed that the predictions of embryonic development fate based on these biomarkers were highly accurate.

The first week following insemination is the period of major pregnancy failure in pasture-grazed dairy cows

D. K. Berg et al.

Summary: The study found that the pregnancy success rate of dairy cows in a seasonal, pasture-grazed system varies at different developmental stages. The major developmental events contributing to pregnancy failure in the first week after fertilization are fertilization failure and embryonic arrest before the morula stage. Pregnancy failure in the second week is primarily associated with embryo elongation failure. Risk factors for pregnancy success related to the cows include the interval between calving and insemination, as well as plasma progesterone concentrations on day 7, while the insemination sire is associated with pregnancy outcome.

Growing cattle embryos beyond Day 8-An investigation of media components

Ekaterina Isaac et al.

Summary: A novel extended culture medium for cattle embryos was developed, allowing growth until embryonic day 12. The inclusion of specific growth factors and additives significantly improved culture success, with Artemin and BMP4 providing the greatest benefit. Replacement of ITSX with B27 led to the most successful embryo culture.

The global gene expression outline of the bovine blastocyst: reflector of environmental conditions and predictor of developmental capacity

Dessie Salilew-Wondim et al.

Summary: The study compared the transcriptome profiles of in vivo and in vitro derived bovine blastocysts, revealing the effects of developmental competence and environmental conditions on gene expression outlines. It also identified differentially expressed genes and pathways reflective of developmental capacities, suggesting that transcriptome activity in blastocysts may indicate future success in pregnancy and adaptation to culture environment.

Transcriptome of D14 in vivo x in vitro bovine embryos: is there any difference?

Ligiane Oliveira Leme et al.

Summary: The study revealed that in vitro culture impacts the transcriptional profiles of bovine embryos, with trophoblast cells from in vitro embryos still showing differences compared to in vivo embryos. Differential gene expression in some key genes may contribute to embryonic losses during the early stages of gestation.

Application of multi-omics data integration and machine learning approaches to identify epigenetic and transcriptomic differences between in vitro and in vivo produced bovine embryos

Maria B. Rabaglino et al.

Summary: By integrating multi-omics data, differentially expressed and methylated genes between in vitro produced (IVP) embryos and embryos produced through ovarian superovulation (MOET) were identified, impacting trophoblast function. Early conceptuses from IVP embryos exhibit epigenetic and transcriptomic changes affecting elongation and focal adhesion, potentially impairing post-transfer survival.
