In vitro culture of ovine embryos up to early gastrulating stages

Priscila Ramos-Ibeas et al.

Summary: Developmental failures occurring after blastocyst hatching from the zona pellucida are a major cause of pregnancy losses in both humans and farm ungulates. Current culture systems cannot support ungulate embryo development beyond hatching, limiting our understanding of key processes for embryo survival. This study reports a culture system that recapitulates most developmental landmarks of gastrulating ovine embryos, providing a valuable platform for studying pregnancy failures during the second week of gestation.

Exclusive generation of rat spermatozoa in sterile mice utilizing blastocyst complementation with pluripotent stem cells

Joel Zvick et al.

Summary: This study successfully achieved complementation of functional spermatozoa in adult chimeras through the technique of blastocyst complementation. Using mouse and rat pluripotent stem cells, the researchers were able to generate chimeras with either species-specific sperm or interspecies sperm capable of fertilization. Single-cell RNA sequencing was used to study spermatogenesis in the chimeric testis. The findings of this study have important implications for germ cell research and conservation efforts for endangered animal species.

Summary: The study reveals that formative stem (FS) cells, a novel type of stem cells, can directly respond to germ cell induction and differentiate into somatic tissues and germline in chimeras. Unlike embryonic stem cells (ESCs) or epiblast-derived stem cells (EpiSCs), FS cells exhibit distinct transcription factor dependencies, primarily relying on Otx2.

Masaki Kinoshita et al.

Summary: This study successfully established pluripotent stem cell lines from porcine, ovine, and bovine embryos without using feeder cells or serum factors. The cells showed differentiation in teratomas and in vitro, with global transcriptome analyses revealing commonality in transcription factor expression across the three species. The research also demonstrated the generation of cloned porcine fetuses using genetically modified AFX stem cells through nuclear transfer.

Summary: Cell competition is a fitness-sensing process where fitter cells eliminate less-fit but viable cells. The study developed an interspecies PSC co-culture strategy and identified a previously unknown mode of cell competition between species. Genetic inactivation of key components of the NF-kappa B complex in human cells improved the survival and chimerism of human cells in early mouse embryos.
