Boosting Endogenous Copper(I) for Biologically Safe and Efficient Bioorthogonal Catalysis via Self-Adaptive Metal-Organic Frameworks

Summary: In order to boost the endogenous Cu(I) level for intracellular drug synthesis through the bioorthogonal reaction, a self-adaptive bioorthogonal catalysis system was constructed by encapsulating prodrugs and sodium ascorbate within adenosine triphosphate aptamer-functionalized metal-organic framework nanoparticles. The system presents specificity to tumor cells and does not require exogenous Cu catalysts, thereby leading to high anti-tumor efficacy and minimal side effects both in vitro and in vivo. This work will open up a new opportunity for developing biosafe and high-performance bioorthogonal catalysis systems.

Liquid Metal as Bioinspired and Unusual Modulator in Bioorthogonal Catalysis for Tumor Inhibition Therapy

Summary: Bioorthogonal catalysis mediated by Pd-based transition metal catalysts has gained interest in combating diseases, but the current Pd-0 catalysts are not efficient enough. In this study, a LM-Pd catalyst with liquid metal as an unusual modulator has been designed for efficient bioorthogonal catalysis for tumor inhibition. The LM matrix stabilizes the active Pd-0 and promotes the uncaging process, while the photothermal properties of LM enhance the removal of tumor cells. This work expands the application of LM and inspires the design of bioinspired bioorthogonal catalysts.

Supported Anionic Gold Nanoparticle Catalysts Modified Using Highly Negatively Charged Multivacant Polyoxometalates

Summary: A feasible method was developed for preparing supported anionic gold nanoparticle catalysts using multivacant lacunary polyoxometalates, confirming the strong electronic interaction between gold nanoparticles and the polyoxometalates, allowing for sequential modulation of the electronic states of the catalysts. The catalyst prepared using [SiW9O34](10-) showed superior catalytic performance and stability compared to supported gold nanoparticle catalysts without this modification.

pH sensing controls tissue inflammation by modulating cellular metabolism and endo-lysosomal function of immune cells

Summary: This study investigates the role of the pH sensor GPR65 and its Ile231Leu variant in inflammation control. The results show that the GPR65 Ile231Leu variant leads to cytokine imbalance, abnormal T cell differentiation, and impaired IL-22 production, making mice highly susceptible to bacterial infection-induced colitis and T cell-driven colitis. Additionally, in dendritic cells, GPR65 Ile231Leu alters endo-lysosomal fusion and degradation capacity, enhancing antigen presentation. This study highlights the importance of pH sensing in inflammatory circuits and tissue homeostasis.

Structural Transformative Antioxidants for Dual-Responsive Anti-Inflammatory Delivery and Photoacoustic Inflammation Imaging

These inhibitors also disrupt bacterial tolerance, reduce biofilm formation, and decrease the number of persister bacteria surviving antibiotic treatment.
