Thioester-mediated biocatalytic amide bond synthesis with in situ thiol recycling

Summary: The activation of carboxylic acids to thioesters, especially Coenzyme A-based ones, is important but challenging. A generic thioester recycling enzyme, CARsr-A, was developed to catalyze the conversion of carboxylic acids to thioesters. This recycling system can regenerate thioesters for various acyl-S-Coenzyme A-dependent enzymes and enable the formation of diverse amides and non-native acylation reactions.

Thiolases can be classified into subfamilies based on unique sequence fingerprints and have distinctive active site properties. This article reviews recent research on the reaction mechanism of thiolases and discusses remaining questions.

This tool is of great significance in predicting the functions of uncharacterized enzymes.

Enzymatic Fluoromethylation Enabled by the S-Adenosylmethionine Analog Te- Adenosyl-L-(fluoromethyl)homotellurocysteine

Summary: Fluoromethyl, difluoromethyl, and trifluoromethyl groups are important in pharmaceuticals and agrochemicals for their roles in efficacy and metabolic stability. Late-stage incorporation of fluorine-containing atoms in molecules has become a significant field in organic and medicinal chemistry, as well as synthetic biology. This study presents a novel fluoromethylating agent, FMeTeSAM, which is structurally and chemically similar to S-adenosyl-L-methionine (SAM) and supports efficient transfer of fluoromethyl groups to oxygen, nitrogen, sulfur, and some carbon nucleophiles. FMeTeSAM is also applied in the fluoromethylation of precursors to oxaline and daunorubicin, two natural products with antitumor properties.

Summary: SAM-dependent methyltransferases (MTs) and SAM analogs can be used for biocatalytic alkylation reactions with high selectivity. Halide methyltransferases (HMTs) enable the synthesis and recycling of SAM analogs using easily available haloalkanes. Anion MTs show promiscuity towards alkyl chains and halide leaving groups, and they cluster in sequence space. This study expands the application of SAM analogs in regioselective reactions using haloalkanes as substrates.

Enzymatic Nitrogen Insertion into Unactivated C-H Bonds

Summary: In this study, new-to-nature heme-containing nitrene transferases were used as starting points for the directed evolution of enzymes to selectively aminate and amidate unactivated C(sp3)-H sites. The evolved enzymes showed high promiscuity and activity towards a wide array of substrates, providing a foundation for further evolution of nitrene transferase function. Computational studies and kinetic isotope effects supported a stepwise radical pathway. In-enzyme molecular dynamics simulations revealed a predominantly hydrophobic pocket with favorable interactions with the substrate.

On the evolution of coenzyme biosynthesis

Identification of an α-Oxoamine Synthase and a One-Pot Two-Step Enzymatic Synthesis of α-Amino Ketones

Summary: Alb29 is an alpha-oxoamine synthase involved in albogrisin biosynthesis in Streptomyces albogriseolus MGR072, responsible for incorporating L-glutamate into acyl-coenzyme A substrates. A one-pot enzymatic system was established to synthesize seven alpha-amino ketones by combining Alb29 with Mgr36 (an acyl-coenzyme A ligase). When these alpha-amino ketones were fed into the alb29 knockout strain Delta alb29, albogrisin analogs with different side chains were observed.
