Advances in Escherichia coli Nissle 1917 as a customizable drug delivery system for disease treatment and diagnosis strategies

Summary: With the advancement in the study of bacteria within the human body, bacterial-based drug delivery system has emerged as a biomimetic platform that can retain biological functions. Escherichia coli Nissle 1917 (ECN) has gained attention as a biological carrier in the treatment of inflammatory bowel disease and tumor due to its biocompatibility and targeting ability. ECN acts as a natural capsule and can be modified or genetically engineered to enhance its function and survival in harsh environments. This review discusses the properties of ECN, its relationship with inflammation and tumor, and strategies to modify it as a delivery carrier for disease treatment.

Structurally Simple Osmium(II) Polypyridyl Complexes as Photosensitizers for Photodynamic Therapy in the Near Infrared

Summary: Five osmium(II) polypyridyl complexes with different ligands were synthesized as photosensitizers for photodynamic therapy. These complexes exhibited high absorption in the near-infrared region and showed promising phototoxicity against cancer cells in vitro and in multicellular tumor spheroids upon irradiation at 740 nm. The complex with 2,2'-bipyridine ligand showed the highest phototoxicity against various cancer cell lines. Experiments on tumor-bearing mice also demonstrated significant tumor growth reduction with laser irradiation at 740 nm. This structurally simple complex has great potential as a photosensitizer for cancer treatment.

A Dual-Metal-Catalyzed Sequential Cascade Reaction in an Engineered Protein Cage

Summary: In this study, an artificial protein cage was created to house a dual-metal-tagged guest protein. The guest protein, consisting of a fusion protein of HaloTag and monomeric rhizavidin, catalyzed a linear, two-step sequential cascade reaction. The protein capsid encapsulated a ruthenium-catalyzed deprotection reaction followed by a gold-catalyzed hydroamination reaction, resulting in the production of indoles and phenanthridines with up to 66% overall yield in aqueous solutions. Additionally, the encapsulation of the guest protein stabilized the metal catalysts against deactivation by air, proteins, and cell lysate.

Near-infrared-activated anticancer platinum(IV) complexes directly photooxidize biomolecules in an oxygen-independent manner

Summary: This study reports the use of near-infrared-activated small-molecule Pt(IV) photooxidants to directly oxidize intracellular biomolecules. These photooxidants can effectively eliminate cancer stem cells and show low toxicity in the dark. Metal-enhanced photooxidation induced by these Pt(IV) complexes leads to robust photooxidation of survival-related biomolecules, inducing oxidative stress, disrupting intracellular pH (pHi) homeostasis, and initiating nonclassical necrosis.
