Synthetic High-Density Lipoprotein-Based Nanomedicine to Silence SOCS1 in Tumor Microenvironment and Trigger Antitumor Immunity against Glioma

Manipulating Neovasculature-Targeting Capability of Biomimetic Nanodiscs for Synergistic Photoactivatable Tumor Infarction and Chemotherapy

Summary: Tumor infarction therapy has potential as an effective antitumor strategy, but its clinical application is hindered by tumor recurrence and nonspecific vasculature targeting. In this study, a neovascular-targeting high-density lipoprotein nanodisc loaded with pyropheophorbide-a and camptothecin was developed for photoactivatable tumor infarction and synergistic chemotherapy. The plane-modified nanodiscs showed higher cellular uptake and could induce thrombus formation upon laser irradiation, while releasing camptothecin to kill residual tumor cells and inhibit recurrence. These findings demonstrate the potential of plane-modified sHDL nanodiscs as versatile drug delivery nanocarriers.
