Summary: Asymmetric reactions that convert racemic mixtures into enantioenriched amines, especially those with a tetrasubstituted carbon stereocenter, are of great importance in pharmaceutical research. In this study, a chiral diene-ligated rhodium-catalyzed asymmetric substitution reaction was developed to efficiently prepare α-trisubstituted-α-tertiary amines containing pharmaceutically relevant secondary amine cores.

Dynamic Kinetic Resolution of Indole-Based Sulfenylated Heterobiaryls by Rhodium-Catalyzed Atroposelective Reductive Aldol Reaction

Summary: A highly enantio- and diastereoselective dynamic kinetic resolution (DKR) of 3-aryl indole-2-carbaldehydes is achieved via a Rh-catalyzed intermolecular asymmetric reductive aldol reaction. The labilization of the stereogenic axis is facilitated by a transient Lewis acid-base interaction (LABI) between the formyl group and a thioether moiety located at the ortho position. The resulting atropisomeric indole products possess a high degree of functionalization and can be further utilized in drug discovery and asymmetric catalysis.

Non-Linear Effects in Asymmetric Catalysis: Impact of Catalyst Precipitation

Summary: Non-linear effects between enantiomeric excesses of ligand and product are common in asymmetric catalysis, and they are useful tools for mechanistic investigations. This study shows that catalyst precipitation affects the shape and amplitude of non-linear effects curves, and it also observes several peculiar features. Overall, this research provides a better understanding of chiral catalytic systems.

Absence of Non-Linear Effects Despite Evidence for Catalyst Aggregation

Rhodium-Catalyzed Regio- and Enantioselective Allylic Amination of Racemic 1,2-Disubstituted Allylic Phosphates

Summary: In this study, alkynylphosphines were utilized as ligands in asymmetric metal catalysis for highly regio- and enantioselective allylic amination reactions. Chiral bis(oxazoline)alkynylphosphine ligands were synthesized and successfully employed in Rh-catalyzed reactions to produce chiral 1,2-disubstituted allylic amines with high yields and excellent enantioselectivity. The presence of a smaller linear alkynyl group on the phosphorus atom in the ligands played a crucial role in accommodating bulky 2-R' groups.
