Antimicrobial Transformation Products in the Aquatic Environment: Global Occurrence, Ecotoxicological Risks, and Potential of Antibiotic Resistance

Paul Loffler et al.

Summary: This study reviews the antimicrobial transformation products (TPs) in surface water environments and evaluates their potential for promoting antimicrobial resistance (AMR) and their ecological and health hazards. Certain TPs were found to pose a risk of AMR promotion and harm to ecosystems and human health. The study proposes a method for evaluating TP risk based on structural similarity and identifies 13 high-priority TPs.

Machine learning-assisted data filtering and QSAR models for prediction of chemical acute toxicity on rat and mouse

Tao Bo et al.

Summary: Machine learning methods can be used to build QSAR models for predicting chemicals' toxicity. However, the model robustness is limited due to poor data set quality for certain chemical structures. To address this issue, a large data set on rat oral acute toxicity was built and machine learning was used to filter chemicals favorable for regression models. The proposed strategy greatly improved the performance of regression models and achieved satisfactory results in classification models.

Occurrence and risks of 23 tire additives and their transformation products in an urban water system

Hai-Yan Zhang et al.

Summary: The presence of tire additives and their transformation products in the urban water system of the Pearl River Delta region in South China was investigated. Nineteen target compounds were detected in surface runoff, with 1,3-Diphenylguanidine (DPG) showing the highest concentration. Benzothiazole and its transformation products were detected at a frequency of 100%, and the antioxidant derivative N-(1,3-dimethylbutyl)-N '-phenyl-p-phenylenediamine-quinone (6PPD-Q) was also found. WWTPs influents and effluents showed the presence of eleven and eight compounds respectively. The study highlights the ecological risks of certain compounds in surface waters and estimates the human daily intake of tire additives.

Pharmaceutical pollution of the world's rivers

John L. Wilkinson et al.

Summary: Environmental exposure to active pharmaceutical ingredients (APIs) can have negative effects on ecosystems and human health. A global-scale study of API pollution in rivers highlights the highest concentrations in sub-Saharan Africa, south Asia, and South America, primarily in low- to middle-income countries with poor wastewater and waste management infrastructure. The most frequently detected APIs include carbamazepine, metformin, and caffeine, with some concentrations exceeding safe levels for aquatic organisms. Pharmaceutical pollution poses a global threat to environmental and human health and has implications for achieving the United Nations Sustainable Development Goals.

Conversion of oxybenzone sunscreen to phototoxic glucoside conjugates by sea anemones and corals

Djordje Vuckovic et al.

Summary: The reported toxicity of oxybenzone-based sunscreens to corals has raised concerns about the impacts of ecotourist-shed sunscreens on weakened corals. The researchers found that oxybenzone caused high mortality of sea anemone under simulated sunlight including UV radiation. Understanding metabolite phototoxicity should facilitate the development of coral-safe products.

A QSAR-ICE-SSD model prediction of the PNECs for alkylphenol substances and application in ecological risk assessment for rivers of a megacity

Yajun Hong et al.

Summary: This study focuses on developing computational models to predict the aquatic toxicity of alkylphenols (APs) with incomplete/unknown toxicity data in aquatic environments. A QSAR-ICE-SSD model was constructed for aquatic organisms, combining quantitative structure-activity relationship (QSAR), interspecies correlation estimation (ICE), and species sensitivity distribution (SSD) models to obtain the hazardous concentrations (HCs) of selected APs.

Screening of Endocrine Disrupting Potential of Surface Waters via an Affinity-Based Biosensor in a Rural Community in the Yellow River Basin, China

Jisui Tan et al.

Summary: In this study, a portable nuclear receptor-based biosensor was developed for highly sensitive analysis of a wide range of potential endocrine-disrupting chemicals (EDCs) in environmental water samples. The biosensor showed a detection limit of 5 ng/L E-2-binding activity equivalent (E-2-BAE) and 93 ng/L T3-BAE. The biosensor was used to map the estrogenic binding activities of surface waters from a rural community in the Yellow River basin in China, and a high correlation was observed when compared with the traditional yeast two-hybrid bioassay.

Joint structural annotation of small molecules using liquid chromatography retention order and tandem mass spectrometry data

Eric Bach et al.

Summary: Structural annotation of small molecules in biological samples remains a challenge in untargeted metabolomics. In this study, a machine learning framework called LC-MS(2)Struct is proposed to improve the annotation accuracy by combining state-of-the-art MS2 scorers and observed retention orders. The method demonstrates significantly higher annotation accuracy and highlights the importance of stereochemistry-aware molecular fingerprints for future LC-MS2 developments.
