By using a simple unimolecular dual-labeled iM scaffold and modifying it with abasic or alkyl linkers, the thermal stability and pH-induced conformational transitions of the iM were improved.

pH-Dependent Capping Interactions Induce Large-Scale Structural Transitions in i-Motifs

Summary: We studied a DNA oligonucleotide that forms two different i-motif structures depending on pH and temperature. At neutral pH, the major structure is stabilized by C:C+ base pairs and G:C:G:C tetrads. At pH 5, a more elongated i-motif structure with C:C+ base pairs and G:T:G:T tetrads is observed. Molecular dynamics calculations showed that the conformational transition between the two structures is driven by the protonation state of key cytosines. This study reveals the pH-dependent plasticity and conformational switch of i-motif structures.

Separation and characterization of bovine milk proteins by capillary electrophoresis-mass spectrometry

Summary: The protein profiling of whey and casein proteins in bovine milk is important in food science and technology. This study presents a novel and simple capillary electrophoresis-mass spectrometry method for the separation and identification of these proteins within 15 minutes. High-resolution mass spectrometry is shown to be valuable in characterizing different protein proteoforms in bovine milk accurately.

The results reveal that the i-motif structures in the promoters of BCL2 and KRAS are highly dynamic and exist in pH-dependent equilibrium with hairpin and hybrid i-motif/hairpin species. The study provides valuable insights into the factors that influence the molecular-level equilibrium between different conformational species involved in oncogene expression.

Thermal and pH Stabilities of i-DNA: Confronting in vitro Experiments with Models and In-Cell NMR Data

Summary: Recent studies have shown that i-DNA, a four-stranded cytosine-rich DNA known as the i-motif, is formed in vivo. This study tested 271 different sequences and found that i-DNA stability is nearly independent on total spacer length, with the central spacer playing a special role in stability. The study also revealed that stability depends on the length of the C-tracts at both acidic and neutral pHs.
