Rapid and high-throughput testing of antifungal susceptibility using an AIEgen-based analytical system

Xiaoxue Ge et al.

Summary: Increasing resistance among fungi to antimicrobials poses global health threats. A new antifungal susceptibility testing (AFST) method using a luminogen with aggregation-induced emission characteristics (AIEgen) called DMASP can assess fungal susceptibility to various antimicrobials within 12 hours, rapidly detect possible drug-resistant fungal strains, and guide the precise use of antimicrobials.

Recognition of Diagnostic Gaps for Laboratory Diagnosis of Fungal Diseases: Expert Opinion from the Fungal Diagnostics Laboratories Consortium (FDLC) COMMENT

Sean X. Zhang et al.

Summary: Fungal infections pose a growing threat to immunocompromised and other medically complex patients, yet there has been little progress in improving fungal diagnostics. The newly established FDLC aims to address diagnostic gaps, with identified shortcomings including lack of molecular detection methods, suboptimal diagnostic algorithms, and inadequate capacity for point-of-care testing for emerging fungal pathogens.

Polarization-sensitive stimulated Raman scattering imaging resolves amphotericin B orientation in Candida membrane

Pu-Ting Dong et al.

Summary: This study used polarization-sensitive stimulated Raman scattering to visualize the distribution of AmB, ergosterol, and lipid in single fungal cells, revealing intracellular lipid droplet accumulation in response to AmB treatment. AmB was primarily located in the cell membrane and intracellular droplets, supporting the ion channel model.

Development of a Fast Raman-Assisted Antibiotic Susceptibility Test (FRAST) for the Antibiotic Resistance Analysis of Clinical Urine and Blood Samples

Xiaofei Yi et al.

Summary: Human health is facing great risks due to the spread of antimicrobial resistance, which has led to the development of a fast Raman-assisted antibiotic susceptibility test (FRAST). This new method showed an overall agreement of 88.0% with conventional AST, and can further reduce turnaround time by using a gram-staining classification based on linear discriminant analysis of SCRS. By applying FRAST to real clinical analysis, it provides fast and reliable susceptibility tests which can speed up microbiological analysis for clinical practice and facilitate antibiotic stewardship.

Recent Development of Rapid Antimicrobial Susceptibility Testing Methods through Metabolic Profiling of Bacteria

Chen Chen et al.

Summary: Inappropriate use of antibiotics leads to the rise of antibiotic-resistant bacteria, necessitating rapid bacterial susceptibility testing. Current reliable phenotypic methods are too slow, calling for new rapid AST methods.

Rapid antimicrobial susceptibility testing by stimulated Raman scattering metabolic imaging and morphological deformation of bacteria

Weifeng Zhang et al.

Summary: A new method based on SRS microscopy was reported to rapidly determine the antimicrobial susceptibility of beta-lactam antibiotics by measuring both the metabolic activity and morphological deformation of bacteria. The evaluation method achieved a 93.2% categorical agreement and a 93.2% essential agreement with the standard reference method when performing AST of cefotaxime on 103 E. coli strains.

Multi-color stimulated Raman scattering with a frame-to-frame wavelength-tunable fiber-based light source

Thomas Wuerthwein et al.

Summary: The study introduces a multi-color imaging technique using stimulated Raman scattering (SRS) with an ultrafast fiber-based light source, featuring integrated amplitude modulation and frame-to-frame wavelength tuning. The light source exhibits low noise level and fast tunability, making it suitable for high chemical specificity imaging assessments.
