Renal-Clearable Probe with Water Solubility and Photostability for Biomarker-Activatable Detection of Acute Kidney Injuries via NIR-II Fluorescence and Optoacoustic Imaging

Fang Zeng et al.

Summary: Acute kidney injuries (AKI) have serious health threats and developing high-performance NIR-II probes for noninvasive detection of AKI is of great significance. In this study, a renal clearable, water soluble, and biomarker activatable probe PEG3-HC-PB was developed for real-time imaging of contrast-agent-induced and ischemia/reperfusion-induced AKI in mice using 3D-MSOT and NIR-II fluorescent dual-mode imaging. This probe can be utilized as a practical tool for detecting AKI and offers insights into the design of other large-conjugation NIR-II probes.

Cisplatin nephrotoxicity: new insights and therapeutic implications

Chengyuan Tang et al.

Summary: The authors review the mechanisms of cisplatin-induced kidney injury and discuss the challenges and potential targets for renoprotection. The cellular and molecular mechanisms of cisplatin nephrotoxicity involve DNA damage, mitochondrial pathology, oxidative stress, endoplasmic reticulum stress, and stress responses such as autophagy, cell-cycle arrest, senescence, apoptosis, programmed necrosis, and inflammation. Emerging evidence suggests a contribution of epigenetic changes to cisplatin-induced kidney injury. Potential therapeutic targets have been identified, but their effects on the efficacy of cisplatin chemotherapy need further evaluation.

Yiteng Ding et al.

Summary: In this study, a reversible fluorescence probe NRN was developed to monitor ONOO???/GSH in an acute kidney injury model. The probe showed excellent selectivity and good sensitivity, and real-time evaluations in vivo were performed. The experiments revealed the severity and irreversibility of acute kidney injury caused by combined drugs under certain conditions. NRN could serve as a potential tool for understanding oxidative stress-related drug-induced acute kidney injury processes.

Rational design of stable heptamethine cyanines and development of a biomarker-activatable probe for detecting acute lung/kidney injuries via NIR-II fluorescence imaging

Juan Ouyang et al.

Summary: Development of high-quality dyes with enhanced NIR-II fluorescence for analyte sensing is crucial for improved imaging depths and resolution. By modifying heptamethine cyanines with pyridinium and tert-butyl groups, steric crowding is increased, water solubility is enhanced, and a site for analyte-responsive elements is provided, resulting in NP-N dyes that outperform benchmark heptamethine cyanines.

In situ detection of alkaline phosphatase in a cisplatin-induced acute kidney injury model with a fluorescent/photoacoustic bimodal molecular probe

Xingwang Chen et al.

Summary: Kidneys play an important role in drug metabolism and excretion. The identification and imaging of effective biomarkers for early evaluation of drug-induced acute kidney injury (AKI) remain significant challenges. Alkaline phosphatase (ALP), a key hydrolyzing protease in the kidney, can be used as a target for testing and imaging of AKI. In this study, a chemically stable molecular sensor (CS-ALP) was developed to map ALP levels in cisplatin-induced AKI, allowing high-fidelity imaging of ALP in AKI.

Early diagnostic imaging of pneumonia with an ultra-sensitive two-photon near-infrared fluorescent probe

Erwang Song et al.

Summary: This study designed and synthesized a two-photon near-infrared fluorescent probe (DCQN) to identify increased concentrations of the inflammation marker SO2 for accurate imaging of pneumonia lesions in deep tissues. The probe specifically reacts with SO2, generating near-infrared fluorescence with good sensitivity, a large Stokes shift, and low detection limit. DCQN has near-infrared emission and good two-photon performance, allowing for the accurate imaging of pneumonia lesions and avoiding interference from background fluorescence from cells.
