A Collection of RPA-Based Photoelectrochemical Assays for the Portable Detection of Multiple Pathogens

Summary: In this study, a photoelectrochemical (PEC) biosensor combined with recombinase polymerase amplification (RPA) technology (RPA-PEC) was proposed for rapid detection of multiple foodborne pathogens. The RPA was initiated on homemade three-dimensional screen-printed paper-based electrodes, forming a lab-on-paper platform. Using the DNA-PEC signaler, photocurrents were achieved after only 20 minutes of RPA, with superior detection performance compared to conventional agarose gel electrophoresis.

CRISPR/Cas12a-powered evanescent wave fluorescence nanobiosensing platform for nucleic acid amplification-free detection of Staphylococcus aureus with multiple signal enhancements

Summary: A CRISPR/Cas12a-powered Evanescent wAve fluorescence nanobiosensing platform (CREAT) was developed for ultrasensitive nucleic acid amplification-free quantitative detection of Staphylococcus aureus (S. aureus) and other pathogens. This nanobiosensor combines several signal enhancement strategies to improve detection sensitivity, including collateral cleavage amplification, nanophotonic structure-based evanescent wave fluorescence enhancement, Mg2+ or DNA-mediated fluorescence enhancement, and air-displacement fluorescence enhancement. The CREAT platform detected S. aureus in real samples with a detection limit comparable to that of RT-qPCR, enabling simple, rapid, sensitive, robust, and flexible on-site detection of S. aureus and other pathogens.

Antibiotic-enzyme-inorganic nanoflowers based immunoassay for the ultrasensitive detection of Staphylococcus aureus

Summary: In this study, a moderate and convenient approach for the determination of staphylococcus aureus (S. aureus) was developed using organic-inorganic hybrid nanoflowers and Pig IgG in an enzyme-linked immunosorbent assay (ELISA) system. The synthesized VAN-HRP-CaHPO4 nanoflowers showed the ability to specifically capture gram-positive bacteria and enhance stability and enzymatic activity for signal amplification. This ELISA system successfully detected S. aureus with a detection limit of 4.3 CFU mL-1, outperforming some commercial ELISA kits. The proposed assay also demonstrated acceptable recovery in detecting S. aureus in food samples.
