Nanopore-based technologies beyond DNA sequencing

Yi-Lun Ying et al.

Summary: This Review discusses the latest advancements in nanopore technologies beyond DNA sequencing. Nanopore techniques have evolved as ultrasensitive analytical tools for individual molecules, enabling applications in molecular sensing and sequencing, chemical catalysis, and biophysical characterization. The prospects of single-protein analysis and sequencing, clinical sensing applications, and synthetic biomimetic nanopores are highlighted. Further exploration of nanopore technologies is expected as control over pore design improves.

Nanopore single-molecule detection of bleomycin via dumbbell DNA scission

Ting Li et al.

Summary: Developing a new method for bleomycin detection using dumbbell DNA as probe on the nanopore sensor platform is described in this study, which shows a quantitative relationship between event intervals (tau(on)) and the concentration of bleomycin with high selectivity and ultra-low detection limit.

Nanopore Sequencing Accurately Identifies the Cisplatin Adduct on DNA

Fubo Ma et al.

Summary: Researchers successfully identified cisplatin lesions by constructing model sequences containing a single cisplatin lesion and using nanopore sequencing (NPS), which can clearly distinguish these lesions on DNA. High-frequency noises caused by cisplatin lesions were consistently observed during NPS, which were not seen in any other combinations of natural DNA bases. Further investigations confirmed the feasibility of NPS in identifying cisplatin lesions at the genomic level.

Detection and Characterization of Single Cisplatin Adducts on DNA by Nanopore Sequencing

Xinjia Zhao et al.

Summary: This study successfully detects and characterizes single cisplatin adducts on DNA at the single-molecule level with single nucleotide spatial resolution, utilizing features such as dwell time, genome anchored current trace, and basecalling accuracy to differentiate these adducts.
