Summary: By taking advantage of target-assisted electrochemically mediated atom transfer radical polymerization (teATRP) for signal amplification, a simple and cost-effective electrochemical aptasensor is developed for sensitive and selective detection of low-abundance endotoxin. The aptamer receptor selectively captures endotoxin, and signal amplification is achieved through the dual amplification strategy of ATRP-initiated polymer chain grafting. This aptasensor shows high sensitivity with a detection limit of 1.2 fg/mL and has great potential for detecting low-abundance endotoxin and other glycan chain containing bio-targets.

Summary: The study found that Pertuzumab undergoes changes in charge heterogeneity, primarily due to deamidation in the Fc domain and N-terminal pyroglutamate formation in the heavy chain. Peptide mapping analysis also showed that clinical samples had 2-3% deamidation in the heavy chain CDR2, 20-25% deamidation in the Fc domain, and 10-15% N-terminal pyroglutamate formation in the heavy chain.

Summary: Antibody combination therapies have emerged as viable treatment options for severe diseases like cancer. However, the co-formulation production approach poses challenges for the analytical control of drug product quality due to complex variant profiles. This study developed a multi-dimensional liquid chromatography with tandem mass spectrometry method to characterize antibody homo- and hetero-aggregates. The results showed that the co-formulation of trastuzumab and pertuzumab, two monoclonal antibodies with similar properties, exhibited high stability with minimal aggregate formation under storage and stress conditions. The formation of homo- and hetero-aggregates was not significantly affected by formulation and storage conditions.

Summary: This research presents a method for rapid quantitation of therapeutic monoclonal antibodies (mAbs) using affinity glass-fiber membranes in a 96-well-plate format. The membrane adsorbs a film containing poly(acrylic acid), allowing the capture of mAbs through covalent-linking. The captured mAbs are then detected using fluorescence intensity, providing a convenient and quick way to quantify mAbs with low variability.

Summary: A novel ratiometric electrochemical biosensor with methylene blue (MB) as the only signal tag was developed for highly reliable and ultrasensitive detection of microRNA-21 (miRNA-21). The strategy utilized an intelligent target-induced dual signal amplification (T-DSA) to controllably load and release MB, avoiding false positive responses. With the aid of innovative T-DSA recycle and rolling circle amplification (RCA)-produced functional DNA nanospheres, the detection sensitivity of this biosensor was significantly improved.

Summary: In this study, functionalized-nanopore-based single-entity electrochemistry analysis tools were used to monitor the single-molecule glycoprotein-boronate affinity interaction. The results showed that the equilibrium association constant of this interaction in a confined nanopore was larger than that in an open system, possibly due to enhanced multivalent synergistic binding. Additionally, the method also demonstrated potential application value for the label-free detection of glycoproteins in real samples, paving the way for single-cell analysis.

Summary: A boronate-affinity cross-linking method is demonstrated for the highly sensitive and selective detection of glycoconjugates. The method utilizes a methylene blue-tagged nucleic acid aptamer to selectively capture target glycoconjugates, followed by labeling a large number of ferrocene tags through boronate-affinity cross-linking. The dual-signal output from the methylene blue and ferrocene tags enables ratiometric detection. This method is simple, cost-effective, and possesses high robustness and reproducibility.
