Native Ion Mobility-Mass Spectrometry-Enabled Fast Structural Interrogation of Labile Protein Surface Modifications at the Intact Protein Level

Gongyu Li et al.

Summary: This study investigates the impact of sialylation on the cytotoxicity of β-amyloid (Aβ) and the stability of transferrin using IM-MS technology. The results show that Neu5Gc substitution enhances Aβ cytotoxicity, while sialylation stabilizes the conformation of transferrin but inhibits its dimerization.

Collision Cross Sections for Native Proteomics: Challenges and Opportunities

Brandon T. Ruotolo

Summary: Recent advancements have made it possible to comprehensively understand protein structure, oligomeric state, sequence, and modification status, providing new possibilities for treating human diseases. Mass spectrometry (MS) tools, especially those associated with ion mobility (IM) separation, have played a crucial role in this progress. However, the application of collision cross section (CCS) values in native proteomics still has limitations.

Approaches to Heterogeneity in Native Mass Spectrometry

Amber D. Rolland et al.

Summary: Native mass spectrometry is a method to preserve and determine the native structure, composition, and stoichiometry of biomolecules and their complexes after they are transferred into the gas phase. Recent advancements in instrumentation and experimental methods have allowed for the analysis of more complex and heterogeneous samples, providing insights into the structure and function of these biomolecules. However, the heterogeneity of the samples poses challenges in assigning ion mass, charge, composition, and structure, which can be addressed using deconvolution algorithms and tools.

Collision-Induced Unfolding Reveals Stability Differences in Infliximab Therapeutics under Native and Heat Stress Conditions

Daniel D. Vallejo et al.

Summary: The study demonstrates the sensitivity of CIU to changes in mAb structure induced by heat stress, with DSC being less sensitive to these alterations. Both instrumental platforms are able to differentiate mAbs, although minor differences between the results obtained on the two instruments were observed. CIU-based heatmaps and classification offer a rapid assessment of the most differentiating charge states for analysis of infliximab, conservatively estimated to improve mAb stability and HOS measurements' efficiency by 30-fold compared to standard DSC tools.
