The role of the enzyme glutathione peroxidase 3 (GPx3) in RV pathogenesis is currently unknown. In this study, GPx3-deficient mice with pulmonary artery banding (PAB) surgery showed worsened RV remodeling and dysfunction compared to wild-type mice.

Summary: Circadian misalignment caused by age, jet lag, and shift work leads to maladaptive health outcomes, including cardiovascular diseases. The understanding of the cardiac circadian clock is limited, hindering the development of therapies to restore it. Exercise has been suggested to reset the circadian clock in peripheral tissues, but it does not alleviate the pathological cardiac remodeling caused by the deletion of core circadian gene Bmal1. However, the deletion of Bmal1 affects systemic circadian output, indicating its critical role in regulating both cardiac and systemic circadian rhythm and function.

Summary: In this study, single-cell RNA sequencing and single T cell receptor sequencing were used to investigate cellular and molecular alterations in cardiac fibrosis, revealing AEBP1 as a crucial regulator in myofibroblasts. A significant infiltration of leukocytes, especially cytotoxic and exhausted CD8(+) T cells and pro-inflammatory CD4(+) T cells, was observed within fibrotic myocardium. Additionally, a subset of tissue-resident macrophages, CXCL8(hi)CCR2(+)HLA-DRhi macrophage, was identified in severely fibrotic areas, interacting with activated endothelial cells via DARC to potentially facilitate leukocyte recruitment and infiltration in human heart failure.

Right versus left ventricular remodeling in heart failure due to chronic volume overload

Summary: In heart failure, the mechanisms of right ventricular dysfunction are poorly understood. This study found that when exposed to chronic volume overload, both the right and left ventricles show similar upregulation of stress and metabolic markers. However, the right ventricle may have a relatively larger response to volume overload compared to the left ventricle, potentially due to concomitant pulmonary hypertension. No evidence supports chamber-specific regulation of protein expression in response to volume overload.
