Inhibition of antiapoptotic BCL-2 proteins with ABT-263 induces fibroblast apoptosis, reversing persistent pulmonary fibrosis

Joseph C. Cooley et al.

Summary: Patients with progressive fibrosing interstitial lung diseases (PF-ILDs) have limited treatment options due to fibroblast resistance to cell death. This resistance can be potentially overcome by targeting the anti-apoptotic proteins of the BCL-2 family. Inhibition of these proteins with ABT-263 showed promising results in inducing fibroblast apoptosis and reducing fibrosis in murine models of PF-ILDs.

Hallmarks of aging: An expanding universe

Carlos Lopez-Otin et al.

Summary: Aging is driven by hallmarks that manifest with age, accelerate aging when accentuated experimentally, and can be decelerated, stopped, or reversed with therapeutic interventions. The twelve proposed hallmarks of aging include genomic instability, telomere attrition, epigenetic alterations, loss of proteostasis, disabled macroautophagy, deregulated nutrient-sensing, mitochondrial dysfunction, cellular senescence, stem cell exhaustion, altered intercellular communication, chronic inflammation, and dysbiosis. These hallmarks are interconnected with each other and with the recently proposed hallmarks of health.

Biomechanical Force and Cellular Stiffness in Lung Fibrosis

Richard S. Nho et al.

Summary: Lung fibrosis is characterized by the accumulation of ECM proteins by apoptosis-resistant fibroblasts. Lung epithelial injury leads to fibroblast recruitment and activation, which are necessary for tissue repair. However, under pathological conditions, a vicious cycle of profibrotic factors and cell-matrix signaling promotes the development of lung fibrosis characterized by increased ECM proteins and changes in matrix properties. Understanding the role of ECM in fibrosis can lead to new therapeutic targets and improved outcomes.

Summary: Based on our findings, the integrin beta 1/AKT cascade primarily acted to induce premature senescence in bladder cancer cells cultured in collagen gel, attenuating chemotherapy-induced apoptosis. Targeting integrin beta 1 might be a feasible approach to eradicate bladder cancer.

Matrix Metalloproteinases and Their Inhibitors in Pulmonary Fibrosis: EMMPRIN/CD147 Comes into Play

Lourdes Chulia-Peris et al.

Summary: Pulmonary fibrosis is a disease characterized by abnormal extracellular matrix deposition, fibroblast to myofibroblast transition, and parenchymal disorganization. The balance between matrix metalloproteinases (MMPs) and their tissue inhibitors (TIMPs) is disrupted in pulmonary fibrosis. CD147, in addition to inducing MMP activity, also plays a key role in fibroblast to myofibroblast transition in this disease.

Summary: This study explored the role of MMP14 in idiopathic pulmonary fibrosis (IPF) and found that its deficiency exacerbates fibrotic injury and affects repair, possibly through its anti-senescent activity. Summary: The guidelines presented in this article aim to assist researchers in selecting and interpreting methods to examine autophagy, as well as to help reviewers provide constructive critiques of reports focused on these processes. It emphasizes that no single assay is perfect for every situation, and multiple techniques may be needed to properly monitor autophagy in different experimental settings. The article also highlights the importance of targeting multiple autophagy-related genes in genetic approaches to block autophagy effectively.
