The Distinct Effects of the Mitochondria-Targeted STAT3 Inhibitors Mitocur-1 and Mitocur-3 on Mast Cell and Mitochondrial Functions

Anastasia N. Pavlyuchenkova et al.

Summary: There is evidence that mitochondria and mitochondrial STAT3 are involved in mast cell activation. Mitocur-1 and Mitocur-3 can reduce antigen-dependent mast cell activation by inhibiting mitochondrial STAT3. The current study investigated the mechanisms of action of these mitocurcuminoids on mast cells and mitochondrial functions. Mitocur-1 and Mitocur-3 both caused mitochondrial fragmentation and increased mitochondrial ROS, but their effects on mitochondrial fission were different.

Protein synthesis, degradation, and energy metabolism in T cell immunity

Julia M. Marchingo et al.

Summary: T cell activation and proliferation lead to significant changes in cell size and molecular content, resulting in an increased demand for energy and amino acids. Protein synthesis, a highly energy- and resource-demanding process, is closely linked to T cell energy production and proteome remodeling. The use of high-resolution mass spectrometry to analyze T cell proteomes can enhance our understanding of the regulation of these processes.

Obesity and Fatty Acids Promote Mitochondrial Translocation of STAT3 Through ROS-Dependent Mechanisms

Rachel Conway et al.

Summary: Obesity promotes metabolic and inflammatory diseases, and one mechanism behind this is the mitochondrial translocation of STAT3, which affects nutrient metabolism and immune response. Cells from obese individuals have higher phosphorylation of STAT3 and mitochondrial accumulation. Fatty acid treatment causes accumulation of mitoSTAT3, and blocking it can prevent obesity and fatty acid-induced production of inflammatory cytokines.

Mitochondrial C1qbp promotes differentiation of effector CD8+ T cells via metabolic-epigenetic reprogramming

Xingyuan Zhai et al.

Summary: C1qbp plays a crucial role in the differentiation of effector CD8(+) T cells by regulating mitochondrial oxidative phosphorylation, affecting levels of acetyl-coenzyme A and fumarate, influencing the methylation status of H3K27, and impacting the transcriptional levels of effector genes.

Integrated analysis of multimodal single-cell data

Yuhan Hao et al.

Summary: The study introduces a weighted-nearest neighbor analysis framework to learn the relative utility of each data type in each cell, enabling integrative analysis of multiple modalities. Applied to a CITE-seq dataset, the method constructs a multimodal reference atlas of the circulating immune system and successfully identifies and validates previously unreported lymphoid subpopulations.
